Background: Pleomorphic giant cell adenocarcinoma (PGCA) of the prostate is a rare, aggressive variant characterized by multinucleated giant cells, sarcomatoid features, and resistance to conventional therapies. Despite its recognition in the WHO 2016 guidelines, the molecular drivers and clinicopathological correlates of PGCA remain poorly characterized. This study presents the first integrative clinicogenomic profiling of PGCA, revealing a novel prognostic gene signature with direct implications for diagnosis and treatment.

Low-dimensional perovskites have been demonstrated repeatedly to improve the performance of perovskite photovoltaic devices in both light-to-electricity conversion efficiency and device durability. In this work, the ionic liquid (IL) 1-ethyl-3-methylimidazolium hydrogen sulfate (EMIMHSO) is innovatively introduced as a capping layer, which interacts with the residual PbI on the 3D perovskite top surface to generate the 1D perovskite, EMIMPbI. By adjusting the concentration of the IL, 1D perovskite formations with distinct morphologies is achieved.

The A35 protein, expressed on the enveloped virion of monkeypox (mpox) virus (MPXV), is essential for viral infection and spread within the host, making it an effective antiviral target. In this study, we demonstrated two human anti-A35 monoclonal antibodies (mAbs) displayed potential protection against MPXV in CAST/EiJ mice and rhesus macaques. Using cryo-electron microscopy, we determined two high-resolution structures of the A35 dimer in complex with the fragment of antigen binding of mAb 975 or mAb 981, revealing detailed interactions at the antigen-antibody interfaces.

Despite their terminally differentiated state, human dermal fibroblasts (HDFs) can undergo osteogenic differentiation in vivo under certain pathological conditions, making them promising candidates for bone tissue engineering-though replicating this in vitro would be difficult. Building on prior findings that low-intensity (5 kV/cm) nanosecond pulsed electric fields (nsPEF) can partially reprogram HDFs toward pluripotency and boost their osteogenic capacity, an in vivo bone regeneration complex was fabricated by encapsulating nsPEF-treated cells in a self-healing hydrogel composed of oxidized hyaluronic acid and hydroxypropyl chitosan. In nude mice, these HDFs produced more robust ectopic bone both subcutaneously and within cranial defects, with significantly higher histological scores than untreated controls.

This study aimed to systematically assess and rank the effects of sensorimotor training (ST), whole-body vibration training (WBVT), neurofunctional training (NT), and balance training (BT) on balance performance in older adults.
Methods: A systematic review and network meta-analysis was conducted following PRISMA guidelines, including 49 randomized controlled trials with a total of 3,028 older adults.

E-cadherin downregulation is an epithelial-mesenchymal transition hallmark canonically attributed to transcriptional repression. Here we delineate a metabolite-driven endocytic route of E-cadherin downregulation in inflammation-associated colorectal cancer (CRC). Specifically, IP kinase-2 (IP6K2), a 5-diphosphoinositol pentakisphosphate (5-IP) synthase upregulated in patients with CRC, is activated via a ROS-Src phosphorylation axis elicited by dextran sulfate sodium (DSS), generating 5-IP around adherens junction (AJ) to promote E-cadherin endocytosis and the transcriptional activities of β-catenin.

Two-dimensional metal-organic frameworks (2D MOFs) have emerged as promising platforms for exploring novel quantum phenomena and tunable electronic functionalities. Here, we investigate π-d orbital hybridization in monolayer M(HAT) (M = Ni, Co, Fe; HAT = 1,4,5,8,9,12-hexaazatriphenylene) frameworks by combining density functional theory (DFT) calculations and scanning tunneling microscopy/spectroscopy (STM/STS) characterization. Despite identical lattice geometries, the Ni-HAT framework exhibits a dispersive, gapless band structure, while the Co- and Fe-HAT frameworks display localized electronic states and semiconducting bandgaps.
