Triple-negative breast cancer (TNBC) remains a refractory subtype of breast cancer due to its resistance to various therapeutic strategies. In this study, we introduce a "brake-release and accelerator-pressing" approach to engineer a microneedle patch embedded with copper-doped Prussian blue nanoparticles (Cu-PB) and the ferroptosis inducer sorafenib (SRF) for raised chemodynamic (CDT)/photothermal (PTT) combination therapy against TNBC. Upon transdermal insertion, the dissolving microneedles swiftly disintegrate and facilitate the release of SRF.

The immunosuppressive niche of the breast cancer microenvironment and the vascular basement membrane significantly hinder the efficacy of nanoparticle-based antitumor immunotherapy delivered via the vascular system. This study describes the design of a puncture-delivered tumor implant, utilizing a photo-cross-linked gelatin methacryloyl hydrogel as the matrix, loaded with CuMnO nanoparticles (CMO NPs) and monomethyl fumarate (MMF), enabling the sustained release of nanoparticles through matrix metalloproteinase-responsive degradation. Mechanistically, CMO NPs induce cuproptosis and immunogenic cell death (ICD) in tumor cells, while synergizing with MMF to trigger substantial mtDNA release into the cytosol.

Diabetic Ulceration (DU) is a main category of nonhealing chronic wounds and tends to be vulnerable and ulcerated repeatedly. Prolonged hyperglycemia microenvironment, together with immunochemotactic response dysfunction, eventually contributes to the refractory wound that is hard to heal. In the chronic DU, there exist obstacles such as diabetic wound bacterial infection, persistent inflammation, suppressed angiogenesis, and reduced growth factor proliferation with decreased hypoxia inducible factor 1 alpha (HIF-1α) expression, and these issues have persisted in hindering the rational and effective wound tissue regeneration.

Methods: We collected data from 16S rDNA amplicon sequencing, untargeted metabolomics, and multimodal brain magnetic resonance imaging from 111 MDD patients and 102 healthy controls. We used multistage linked analyses, including group comparisons, correlation analyses, and mediation analyses, to explore the relationships between the gut microbiome (), fecal metabolome, brain imaging, and behaviors in MDD.

Methods: A total of 102 subjects with symptomatic KOA were included in this study. Coronal magnetic resonance imaging was used to measure the meniscal parameters including the mean distance from medial to lateral meniscal lesions (Mean(MLD)), the mean value of tibial plateau width (Mean(TPW)) and the mean of the relative percentage of the medial to lateral meniscal lesions distance (Mean(RMLD)).

Chronic inflammation impedes the healing of diabetic ulcers (DUs), while P-glycoprotein (P-gp)-mediated drug efflux reduces the efficacy of conventional drug formulations. In this study, a novel liposome-mediated hyaluronic acid (HA) microneedle patch (E-Lipo@A-HMN) was developed for enhanced DUs healing via the transdermal delivery of Acetyl-11-keto-β-boswellic acid (AKBA) and β-elemene (ELE). The findings demonstrate that liposomal encapsulation enhances the solubility and bioavailability of ELE, and HA microneedles improve skin penetration and enable controlled drug release by providing a biodegradable, biocompatible scaffold that improves skin penetration and enables controlled drug release.

Oxidative stress leads to intestinal barrier damage, which induces immune responses to occur and further promotes oxidative stress exacerbating inflammatory bowel disease (IBD). In this work, the multifunctional ZnCeO/Se (ZCSO) nanozyme wrapped with acid-resistant calcium alginate hydrogel designed for oral administration is prepared. The ZCSO nanozyme can promote the activation of the Nrf2 oxidative stress pathway, then significantly improve the efficiency of scavenging reactive oxygen species (ROS) and up-regulate the protein expression of glutathione peroxidase 4 (GPx4), which is closely related to the inhibition of ferroptosis.
