To address the current limitations of DNA-compatible Sonogashira cross-coupling reactions capable of accommodating a broad range of commercially available phenolic building blocks (BBs), an SuFEx-Sonogashira cross-coupling protocol has been developed. This protocol involves the conversion of readily accessible phenolic compounds into the corresponding aryl fluorosulfates within 96-well microplates via a highly efficient liquid-phase SuFEx reaction, followed by Sonogashira cross-coupling with DNA-conjugated terminal alkynes.

Understanding hydrogen bonding and ion-specific interactions in water, sodium sulfate (NaSO), and acetonitrile (ACN) systems remains challenging due to their complex, dynamic nature. Here, Raman spectroscopy is employed to probe hydrogen bonding networks and ion reorganization in NaSO aqueous solutions with different ACN concentrations. The results indicate that, at low ACN concentrations in the ternary solutions, hydrogen bonding between ACN and water molecules disrupts the original hydration structure of the ions, resulting in the formation of small ion clusters via electrostatic interactions.

Background: Gastric cancer is one of the most common cancers worldwide, with its prognosis influenced by factors such as tumor clinical stage, histological type, and the patient's overall health. Recent studies highlight the critical role of lymphatic endothelial cells (LECs) in the tumor microenvironment. Perturbations in LEC function in gastric cancer, marked by aberrant activation or damage, disrupt lymphatic fluid dynamics and impede immune cell infiltration, thereby modulating tumor progression and patient prognosis.

Background: Most RNA-seq datasets harbor genes with extreme expression levels in some samples. Such extreme outliers are usually treated as technical errors and are removed from the data before further statistical analysis. Here we focus on the patterns of such outlier gene expression to investigate whether they provide insights into the underlying biology.

Histone arginine methylation by protein arginine methyltransferases (PRMTs) is crucial for transcriptional regulation and is implicated in cancers. Despite their therapeutic potential, some PRMTs present challenges as drug targets due to their context-dependent activities. Here, we demonstrate that hypoxia triggers the rapid condensation of PRMT2, which is essential for its histone H3R8 asymmetric dimethylation (H3R8me2a) activity.

Tomato brown rugose fruit virus (ToBRFV) overcomes all known tomato resistance genes, including the durable Tm-2, posing a serious threat to global tomato production. Here, we employed in vitro random mutagenesis to evolve the Tm-2 leucine-rich repeat (LRR) domain and screened ∼8,000 variants for gain-of-function mutants capable of recognizing the ToBRFV movement protein (MP) and triggering hypersensitive cell death. We identified five such mutants.
