The human retina exhibits complex cellular heterogeneity which is critical for visual function, yet comprehensive ethnic-specific references are scarce in ophthalmic transcriptomics. The lack of single-cell RNA sequencing (scRNA-seq) data from Asian populations particularly Chinese donors imposes significant limitations in understanding population-specific retinal biology. We constructed the first comprehensive single-cell transcriptomic atlas of the human retina from Chinese donors, generated through high-throughput scRNA-seq of ∼290,000 viable cells obtained from 18 fresh retinal specimens (living donor and post-mortem specimens).

Accurate variant calling is essential for next-generation sequencing (NGS)-based diagnosis of rare diseases, yet most benchmarking studies have focused on standard cell lines or trio-based samples, with limited relevance to sporadic cases. Here, we systematically compared the performance of DeepVariant and GATK HaplotypeCaller in two Chinese cohorts of patients with sporadic epilepsy (EP) and autism spectrum disorder (ASD). DeepVariant exhibited higher precision and sensitivity in detecting single nucleotide variants (SNVs), while GATK showed a distinct advantage in identifying rare variants, which are often key to understanding the genetic basis of rare diseases.

The speech and language rehabilitation are essential to people who have disorders of communication that may occur due to the condition of neurological disorder, developmental delays, or bodily disabilities. With the advent of deep learning, we introduce an improved multimodal rehabilitation pipeline that incorporates audio, video, and text information in order to provide patient-tailored therapy that adapts to the patient. The technique uses a cross-attention fusion multimodal hierarchical transformer architectural model that allows it to jointly design speech acoustics as well as the facial dynamics, lip articulation, and linguistic context.

Hydride superconductors have attracted significant attention, yet achieving superconductivity at ambient pressure remains a key challenge. Here, a family of high-T (superconducting critical temperature, T) hydrides based on the fluorite-type AXH structure, exhibiting thermodynamic and dynamic stability at low to atmospheric pressure, is proposed. Through comprehensive screening of 150 ternary systems, eight stable hydrides below 35 GPa are identified.
