The first asymmetric total synthesis of 5,10-seco-neoansamycin A, a 19-membered cyclic octaketide, was achieved in 20 steps (with several steps being telescoped) and 6.7% overall yield starting from the reported compound, thus leading to the assignment of its absolute configuration. This convergent synthetic approach features late-stage macrolactamization, a judicious application of an asymmetric aldol reaction.

Heat shock protein family A member 4-like (HSPA4L) has been shown to be overexpressed in osteoarthritis (OA) patients, but its role in OA process still unknown. Chondrocytes were stimulated with interleukin-1β (IL-1β) to mimic OA cell model in vitro, and rat was injected with monosodium iodoacetate (MIA) to construct OA rat model in vivo. The expression of HSPA4L, methyltransferase-like 3 (METTL3) and extracellular matrix (ECM)-related markers was examined by qRT-PCR or western blot.

Gastric cancer (GC) is the third leading cause of cancer mortality globally, often presenting with insidious symptoms that lead to late-stage diagnoses, underscoring the critical need for innovative diagnostic and therapeutic strategies. One such avenue is the exploration of ferroptosis, a regulated form of cell death implicated in various pathological conditions and malignancies. In this study, we demonstrate that brucine, an alkaloid derived from Strychnos nux-vomica, exerts significant antitumor effects on GC cells both in vitro and in vivo.

Nanozymes, which possess inherent catalytic properties that are akin to those of natural enzymes, have emerged as promising candidates for biomedical innovation. In this work, we successfully synthesize a Co Cu S nanoflower by the solvothermal and soaking method. Fortunately, through cobalt doping and microstructure design, its morphological structure and active sites have been optimized and adjusted, thus bestowing the Co Cu S nanoflower enhanced peroxidase-mimetic activity.

Background: Bullous Pemphigoid (BP) is caused by a predominantly Th2-mediated attack on the basement membrane by the production of anti-BP180 and anti-BP230 antibodies. Malignant tumors can exacerbate immune disorders through a variety of potential pathways, including pro-inflammatory responses in the tumor microenvironment, cross-immune responses induced by tumor-associated antigens, and the lifting of immunosuppressive states and activation of underlying autoimmune responses after surgery. Alopecia Areata (AA) is an autoimmune disease caused by T-lymphocyte-mediated destruction of the immune privilege of the hair follicle, specifically involving the immune axes of Th1, Th2 and Th17.

Objectives: Bladder cancer is a common malignancy with high incidence and poor prognosis. N-methyladenosine (mA) modification is widely involved in diverse physiological processes, among which the mA recognition protein YTH N-methyladenosine RNA binding protein F2 (YTHDF2) plays a crucial role in bladder cancer progression. This study aims to elucidate the molecular mechanism by which O-linked -acetylglucosamine (O-GlcNAc) modification of YTHDF2 regulates its downstream target, period circadian regulator 1 (), thereby promoting bladder cancer cell proliferation.

The brown planthopper (BPH), Nilaparvata lugens (Stål), is a typical insecticide-induced resurgence rice pest that causes severe damage to rice in Asian countries. Previous studies have shown that the fungicide Jinggangmycin (JGM), used to control rice sheath blight disease, can stimulate BPH fecundity; however, the molecular mechanism remains to be further explored. In this study, based on transcriptomic analysis, we found that the PI3K-Akt signaling pathway was significantly enriched in BPH after feeding on JGM-treated rice, where the NlPR-L and NlABD4-L genes were significantly upregulated.

As the weed Echinochloa phyllopogon has rapidly developed multi- and cross-resistance to several herbicides, we aimed to determine the mechanism underlying penoxsulam resistance in weeds. There was no target mutation in the tested population, and P450 enzyme activity was significantly higher in the penoxsulam-treated resistant population, confirming that non-target-site resistance was dominant. The antioxidant enzyme activity of the resistant population was higher than that of the sensitive population following the application of the penoxsulam and cleared HO faster.

Wendan Decoction (WDD), a classic formula for the expectoration of phlegm, has been acknowledged for the management of chronic obstructive pulmonary disease (COPD). Nevertheless, the therapeutic components and mechanism of WDD in COPD remain elusive. Hence, an analytical strategy for extensive investigation of the constituents in WDD combining UFLC-ESI-Q/TOF-MS and subsequent network pharmacology was conducted, and the appropriate preparation of bio-samples was involved with mechanistic bioanalysis including multi-technique metabolomics and molecular biological means, that pivotal factors such as PTGS2, arachidonic acid (AA) metabolism, and PI3K-AKT signaling pathway were recommended as potential central elements for the anti-COPD effects.
