Peptide-based biosensors are widely used for in vitro detection of protease activity but often suffer from the limited sensitivity, poor accuracy, and incompatibility with point-of-care testing (POCT) devices. Herein, we developed a versatile deoxyribozyme (DNAzyme)-amplified protease-sensing (DP) platform that integrates the positively charged oligopeptides with a negatively charged DNAzyme biocatalyst for highly-sensitive protease detection. The system leverages the electrostatic peptide-DNAzyme interactions to inhibit DNAzyme catalytic activity, which is reactivated upon the protease-triggered peptide hydrolysis, thus enabling an efficient signal amplification via the successive cleavage of DNAzyme substrate.

Hemoperfusion is one of the most effective blood purification techniques to quickly remove bilirubin from the blood of patients with kidney or liver failure. Although numerous adsorbent materials with high adsorption capacity have been developed, their clinical application are still limited due to poor biocompatibility and biosafety issues. Tumor-associated macrophages (TAMs) display remarkable functional heterogeneity, yet the molecular mechanisms driving their diverse phenotypes remain elusive. Using CRISPR screens in primary macrophages, we identified tumor-derived factors, including lactic acid, PGE2, and GM-CSF, as key modulators of TAM polarization. These factors interact synergistically and antagonistically to shape distinct TAM phenotypes that are highly conserved across human cancers.

Background: The apolipoprotein E (ApoE) ε4 allele and type 2 diabetes mellitus (T2DM) are independent risk factors for Alzheimer's disease (AD), the most prevalent neurodegenerative disorder in the elderly. The T2DM patients carrying the ApoE ε4 allele exhibit heightened activation of platelet glycogen synthase kinase-3β (GSK-3β), a key downstream kinase in the insulin signaling pathway, along with more severe cognitive deficits. This observation suggests an intrinsic link between ApoE ε4, GSK-3β, and cognitive dysfunction.

Glioblastoma multiforme (GBM) continues to pose a significant challenge in the field of neuro-oncology primarily because of the limited penetration of therapeutics across the blood-brain barrier (BBB) and the presence of immunosuppressive tumor microenvironments. To address these challenges, a HD-PEG@BM biomimetic nanoplatform (hereinafter referred to as HD-P@BM) is developed that cloaks the near-infrared II photosensitizer HD-PEG (HD-P) inside microglial membranes to enable enhanced BBB penetration and tumor-targeted delivery. In this study, it is found that the microglia-derived membranes enhanced the uptake of nanoparticles by both the glioma cells and tumor-associated microglia.

Primary splenic angiosarcoma is a rare, aggressive tumor, characterized by rapid metastasis and poor prognosis. Herein, we report a case of 18F-FDG and 68Ga-FAPI-04 PET/CT imaging in a 57-year-old man with primary splenic angiosarcoma. 68Ga-FAPI-04 PET/CT presented much higher uptake in the lesions compared with 18F-FDG PET/CT, showing better diagnostic performance.

Epilepsy is a frequently occurring complication in patients with gliomas that substantially impairs their quality of life. The onset of epilepsy in patients with gliomas is driven by multiple mechanisms, including tumour-induced compression of the peripheral neural network, the release of neurotransmitters and inflammatory factors by tumour cells, alterations in the tumour microenvironment, and changes in gene expression. Collectively, these factors contribute to the abnormal excitability of local neurons, ultimately triggering seizures.

Integrin αvβ6, an epithelial-specific integrin, plays a crucial role in the development and prognosis of multiple tumors and has emerged as a potential target for cancer diagnosis and treatment. The novel integrin αvβ6-targeted radiotracer [Ga]Ga-Trivehexin shows excellent targeting specificity and favorable tumor-to-background contrast. In this prospective clinical study (NCT05835570) involving 58 participants with non-small cell lung cancer (NSCLC), [Ga]Ga-Trivehexin and 2-deoxy-2-[F]fluoro-D-glucose ([F]FDG) positron emission tomography/computed tomography (PET/CT) showed perfect diagnostic accuracy (100%, 58/58) for the detection of primary tumors.
