Background: Transbronchial cryobiopsy (TBCB) is generally recommended under intubation or rigid bronchoscopy with general anesthesia. However, some patients with diffuse parenchymal lung disease (DPLD) are unable to tolerate general anesthesia, which limits the widespread adoption of TBCB.
Methods: A total of 37 patients with DPLD who underwent TBCB under conscious sedation without intubation were included in this study.

Dendritic cells (DCs) serve a pivotal and diverse role in the modulation of transplant immunity, wherein alterations in their maturation state influence the balance between promoting immunotolerance and exacerbating inflammatory responses. Recent studies have revealed a spectrum of novel mechanisms through which absent in melanoma 2 (AIM2) regulates the functions of immune cells. However, the impact of AIM2 on the regulatory functions of dendritic cells in alloimmunity has not been thoroughly investigated.

Unlabelled: The role of RNA N6-methyladenosine (m6A) modifications in modulating the immune microenvironment during ischemic stroke (IS) pathogenesis remains poorly characterized. This investigation systematically explores m6A-mediated immune regulation in IS and identifies critical immune-related biomarkers. Transcriptomic profiles from 108 IS samples were analyzed to discern m6A regulatory patterns.

Bronchial mucosa-associated lymphoid tissue is the site of the uncommon malignancy known as mucosa-associated lymphoid tissue (MALT) lymphoma. Due to its lack of distinct clinical signs and imaging characteristics, it is frequently misdiagnosed and underdiagnosed. After receiving unsuccessful treatment for a lung infection or tuberculosis in multiple tertiary care hospitals, we report a male patient who had a solid lesion in his left upper lung for 18 years.

Hepatocellular carcinoma (HCC) remains a leading cause of cancer death worldwide. Sonodynamic therapy (SDT) offers a non-invasive, deep-penetrating approach by using ultrasound to activate sonosensitizers and generate cytotoxic reactive oxygen species (ROS). Yet poor intratumoral delivery and low ROS quantum yields of existing agents have stalled clinical translation.

Background: Individuals with dysphagia typically experience multiple co-occurring symptoms, whose interrelationships can vary in strength. The significance and impact of each symptom within dysphagia also differ. However, the intricate web of connections among these symptoms remains poorly understood, making it challenging to assess the importance of individual symptoms from a relational perspective.

G-Quadruplexes play key roles in tumorigenesis, but detecting them remains challenging. We developed NIRGQ-3, a NIR-II fluorescent probe, with 25-fold fluorescence enhancement and improved sensitivity (limit of detection: 35 nM). It enables high-contrast tumor imaging (SBR = 25), providing a powerful tool to decipher G-quadruplex dynamics and advancing cancer diagnostics.

Osteoarthritis (OA) is a common joint disease, and chondrocyte extracellular matrix (ECM) degradation was closely associated with its progression. This study investigated the regulatory mechanisms of ECM degradation during OA development. A rat model of OA was established by anterior cruciate ligament transection (ACL-T) and interleukin-1 beta (IL-1β)-stimulated rat chondrocytes were used to simulate OA in vitro.

Skeletal stem cells (SSCs) are tissue-specific stem cells characterized by their capacity for self-renewal and their position at the apex of the differentiation hierarchy. They can generate mature bone cell types essential for bone development, maintenance, and repair. Lineage tracing experiments have demonstrated that SSCs reside in the bone marrow, periosteum, and the resting zone of the growth plate.

A diffusely thickened or hard pleura is a special type of macroscopic appearance associated with benign or malignant conditions. Medical thoracoscopy (MT) is the gold standard for pleural pathology, but its diagnostic yield is imperfect. Although cryobiopsy may provide greater and deeper tissue, its impact on the diagnostic yield remains uncertain, and safety concerns persist.

Background And Aims: Cholangiocarcinoma (CCA) is one of the most prevalent primary liver malignancies with increasing incidence and mortality rates, particularly in Southeast Asia. Surgical resection is a primary therapeutic option offered to patients with localised disease. Unfortunately, early disease recurrence is common and robust tools for tailored patient-centric post-operative management and follow-up schemes are highly desired but currently lacking.

Background: Triple-negative breast cancer (TNBC) is an aggressive subtype with limited treatment options, and paclitaxel resistance remains a major therapeutic challenge. Ferroptosis, an iron-dependent cell death mechanism, and microRNAs (miRNAs) like miR-325-3p have emerged as potential regulators of chemoresistance, but their roles in TNBC are poorly understood.
Objective: This study aimed to investigate whether ferroptosis and miR-325-3p modulate paclitaxel resistance in TNBC and to identify the underlying molecular mechanisms.

Fibroblast activation protein α (FAPα) has been reported to be expressed in many carcinoma cells and associated with tumor growth, invasive, and metastasis. However, due to the lack of appropriate fluorescent probes, detecting FAPα with high sensitivity in tumor and clearly delineating tumor margins remains a challenge. Herein, we developed a general design strategy-introducing a positive charge into the fluorophore skeleton-for the construction of FAPα-activated fluorescent probe with enhanced sensitivity.
