A method for both qualitative screening and quantitative determination of 569 pesticides in honeysuckle was developed based on ultrahigh-performance liquid chromatography (UHPLC) coupled to quadrupole-Orbitrap high resolution mass spectrometry and an in-house executable compound database involving the theoretical masses of precursor and fragment ions and retention times. Different workflows were evaluated, validated and compared including Full MS-SIM, Full MS/ddMS, Full MS/AIF and Full MS/DIA. For qualitative analysis, Full MS/DIA showed relatively low screening detection limits (SDLs) for most pesticides, but its MS identification seemed to be not reliable enough.

Fructus Psoraleae (FP) causes cholestatic liver injury; however, its main toxic constituents that are responsible for causing hepatotoxicity remained undetermined in previous studies. In the present study, psoralen and isopsoralen, the two main constituents of FP, were administered orally to rats (80 and 40 mg/kg, respectively) and mice (320 and 160 mg/kg, respectively) for 28 days, followed by biochemical and histopathological examinations to evaluate their hepatotoxicity. The results showed that psoralen and isopsoralen could induce the toxic reactions of liver and other organs in rats, while mice were not sensitive to these two compounds.

Perspectives on Liquid Chromatography-High-Resolution Mass Spectrometry for Pesticide Screening in Foods.

J Agric Food Chem

September 2018

Thermo Fisher Scientific , 355 River Oaks Parkway , San Jose , California 95134 , United States.

This perspective discusses the use of liquid chromatography coupled with high-resolution mass spectrometry (LC-HRMS) for multiresidue analysis of pesticides in foods and agricultural commodities. HRMS has the important distinction and advantage of mass-resolving power and, therefore, requires different concepts, experiments, and guidance for screening, identification, and quantitation of pesticides in complex food matrices over triple quadrupole mass spectrometry. HRMS approaches for pesticide screening, including full-scan experiments in conjunction with tandem mass spectrometry (MS/MS) experiments, are described.
