Background: Mavacamten is commercially approved for use in symptomatic obstructive hypertrophic cardiomyopathy (oHCM) patients. This study evaluated its real-world impact on symptoms, echocardiographic changes, and the need for septal reduction therapy (SRT) in oHCM patients treated at a multi-hospital referral center.

Methods: We included 244 oHCM patients (mean age 64 years, 57% female, 21% with atrial fibrillation, 51% New York Heart Association [NYHA] Class III, 82% on beta-blockers) treated with mavacamten for ≥3 months (171 for ≥12 months).

In this study, we demonstrate an octuple-state sliding ladder ferroelectric behaviour in bilayer GeSe/SnS van der Waals heterostructures. This behaviour arises from compression-modulated interlayer sliding, which induces a series of multi-step phase transitions.

Enhancing robustness of spiking neural networks through retina-like coding and memory-based neurons.

Spiking neural networks (SNNs) are emerging as a promising alternative to traditional artificial neural networks (ANNs), offering advantages such as lower power consumption and biological interpretability. Despite recent progress in training SNNs and their performance in computer vision tasks, there remains a question of SNN robustness to corrupted images in real-world scenarios. To address this problem, we introduce CIFAR10-C and IMAGENET-C datasets from the ANN field as benchmarks and further propose novel methods to improve SNN corruption robustness.

DeepRS: deep neural network-based in-service Rayleigh-scattering monitoring in bidirectional mode-division multiplexing systems.

In bidirectional mode-division multiplexing (Bi-MDM) systems, which have the potential for significantly higher transmission spectral efficiency, the complex interplay between mode-coupling and Rayleigh scattering (RS) exacerbates channel instability, posing significant challenges for dynamic network management. To address this, we propose DeepRS, an innovative deep-learning-based scheme for high-precision, in-service RS noise monitoring. By utilizing deep neural networks (DNNs) to extract features from the filtered frequency amplitude histogram (FFAH) of received signals, it enables the efficient signal-to-RS ratio (SRR) monitoring without disrupting signal traffic.

Van Maldergem syndrome-1 in a patient with central precocious puberty: A case report.

Rationale: Van Maldergem syndrome-1 (VMS-1; OMIM #601390) is a multisystem genetic disease characterized by intellectual disability, craniofacial deformities, skeletal anomalies, and/or other variable malformations. Few cases have been reported to date, posing challenges to the diagnosis and management of this condition.

Patient Concerns: A 7-year-old female Chinese patient presented with a series of developmental defects, including precocious puberty, mild intellectual disability, unusual craniofacial features, mild shortening of the fourth metacarpal bone, and clumsy movements with poor coordination.

Methods: The data included 3587 women from the Jiangsu Birth Cohort (JBC) who conceived through assisted reproductive technology (ART) and had gestational ages of at least 20 weeks.

This case report explores the potential of endoscopic photodynamic therapy (PDT) to downstage locally advanced colon cancer (LACC) and enhance patient quality of life. A young female with obstructive LACC, unresponsive to CapeOX chemotherapy, underwent emergency colostomy followed by dual-route endoscopic PDT. Subsequent neoadjuvant therapy, including TAS-102, targeted agents, and immunotherapy, led to a partial tumor response.

Comprehensive tumor-immune profiling reveals mediators of paradoxical immune sensitivity in sarcomatoid renal cell carcinoma.

Renal cell carcinoma with sarcomatoid features (sRCC) is a highly aggressive tumor type yet preferentially responds to immune checkpoint blockade (ICB). To better understand microenvironmental mediators of this paradoxical immune sensitivity, we performed single-cell analyses of human sRCC tumors compared against clear cell RCC (ccRCC), with validation spatially and in bulk transcriptomic datasets totaling over 3,000 RCC tumors. We describe a robust immune network in sRCC using these orthogonal approaches: tumor-infiltrating T cells in sRCC are more activated, and subsequently exhausted, while being enriched for CXCL13 expression.
