In oxaliplatin-resistant gastric cancer (GC), multi-omics profiling combined with organoid libraries reveals altered metabolic pathways associated with chemoresistance. We identify a novel lactylation modification at K115 of Poly(RC)-binding protein 2 (PCBP2K115la), which confers functional oxaliplatin resistance. Mechanistic studies demonstrate that the long non-coding RNA BASP1-AS1 assembles a complex containing Unc-51 Like Autophagy Activating Kinase 1 (ULK1) and lactate dehydrogenase A (LDHA), thereby activating LDHA enzymatic activity to increase lactate production.

Hepatocellular carcinoma (HCC) is notorious for its high likelihood of recurrence even after radical surgery, which calls for effective adjuvant therapy based on more precise patient selection. The decline of the abundance of binuclear hepatocytes (ABH) in paracancerous liver tissues has been reported to indicate pathological changes in liver cells, leading to short-term recurrence within 2 years. In this research, we analyzed 34 HCC patients and 22 patients underwent liver surgery for non-HCC diseases.

We introduce a protocol for generating 3D organoids from circulating tumor cells (CTCs), enabling longitudinal functional and molecular analyses in pancreatic cancer patients, including those with unresectable disease, which constitutes the majority of cases. We outline the process for isolating and characterizing CTCs from the blood of pancreatic cancer patients and provide detailed instructions for initiating, passaging, and phenotyping CTC-derived organoids. Additionally, we describe techniques for utilizing these organoids in drug screening with a focus on stemness-related pathways.
