Glioblastoma (GBM) remains one of the most aggressive and lethal forms of brain cancer, characterised by profound genetic, epigenetic, and phenotypic heterogeneity. Recent advancements in high-resolution genome mapping have unveiled the critical role of three-dimensional (3D) chromatin architecture-encompassing chromatin loops, topologically associating domains, and enhancer-promoter interactions-in driving GBM tumourigenesis and therapy resistance. This review summarises recent insights into the mechanistic contribution of 3D genome reorganisation in sustaining oncogenic transcriptional programs, promoting intratumoural heterogeneity, and facilitating adaptive resistance.

Neutrophils are granular and polymorphonuclear cells and one of the main participants of the innate immune system, which have received considerable attention due to the discovery of neutrophil extracellular traps (NETs). Extracellular vesicles (EVs), particularly those released by immune cells such as neutrophils, have been associated with the immunopathogenesis of autoimmune diseases. Besides, studies have reported a fundamental correlation between EVs and NETosis in autoimmune diseases.

N-methyl-d-aspartate (NMDA) receptors are validated druggable targets for the treatment of Alzheimer's and other associated neurological conditions, particularly in individuals with disabilities. Considering the excitotoxicity associated with NMDA receptors, which leads to neuronal damage, cognitive impairment, and limitations of current therapeutic regimens, better therapeutic candidates are required. One of the validated drug discovery approaches is computer-assisted drug discovery, supplemented by molecular docking, mechanics, and dynamics.

Autoimmune diseases occur when the immune system mistakenly attacks the body's own tissues, affecting millions of people and often requiring long-term treatment. Current therapies, such as immunosuppressants and biologics, help manage symptoms but can cause serious side effects. A promising new approach involves engineered microbiota-a method that modifies gut bacteria to influence immune function and potentially ease autoimmune conditions.

Conventional TiO₂ nanoparticle syntheses rely on high temperatures, toxic reagents and multi-step routes that impede scalability and sustainability. Here, we deliver the first green synthesis of TiO₂ nanoparticles (TiO₂ NPs) using polysaccharide- (42 mg GE g) and phenolic-rich (78 mg GAE g) Pinus patula leaf extract. GC-MS and LC-MS fingerprinting identify terpenoids, flavonoids and phenolic glycosides acting as simultaneous reducing, capping and stabilizing agents.

Alcoholic liver disorder (ALD) is one of the most prevalent hepatic ailments worldwide, with oxidative stress and inflammation playing a vital role in disease progression. The current study intended to assess the anti-inflammatory nature of Hamamelitannin (HAM), a gallotannin from Hamamelis virginiana barks, which was predicted to possess anti-inflammatory properties based on in-silico docking analysis. To further explore its effects, we examined the therapeutic effect of HAM against ethanol-mediated inflammation using an in-vivo zebrafish larvae model.

Quercetin, a naturally occurring flavonoid, has been found to be a potential agent for the treatment of major depressive disorder (MDD), given the complexity of its interaction with the serotonin transporter (SERT). Clinically, quercetin has direct and indirect modulatory effects as opposed to conventional selective serotonin reuptake inhibitors (SSRI), which act mainly by inhibiting SERT after a time delay and communicate with SERT through possible binding location preferences and allosteric processing, while simultaneously controlling its definite expression through anti-inflammatory and antioxidant pathways, such as the NF-kB, AMPK/SIRT-1, and Nrf2-ARE cascades. These processes assist in modifying serotonergic neurotransmission and minimizing oxidative and inflammatory strains, which are the major contributors to the pathophysiology of depression.

Epigenetic modulation has emerged as a central strategy that can change the fate of tumour cells to offer more rational and precise approaches by modulating reversible changes in chromatin structure, regulating gene expression without altering DNA sequence. Many reports have indicated the contributions of abnormal epigenetic alterations, particularly DNA methylation and histone modification patterns, as well as their association with non-coding RNA interactions during cancer emergence, development or resistance to standard therapies. Ongoing studies on various inhibitors also demonstrate encouraging preclinical results and potent inhibitory activity.
