Since the outbreak of SARS-CoV-2 in late 2019, the cumulative number of confirmed cases worldwide has surpassed 778 million, and the number of deaths has exceeded 7 million, posing a significant threat to human life and health while inflicting enormous losses on the global economy. At the stage where sequential immunization is recommended, there is a pressing demand for mRNA vaccines that can be rapidly adapted to new sequences, are easy to industrialize, and exhibit high safety and effectiveness. We developed a lipid nanoparticle (LNP) system, designated as WNP, which facilitates essentially in situ expression at the injection site and results in lower levels of pro-inflammatory factors in the liver, thus enhancing its safety compared to liver-targeted alternatives.

Background And Purpose: Siguan points, comprising bilateral Hegu (LI4) and Taichong (LR3), have shown good therapeutic potential in migraine management. We aimed to systematically evaluate the clinical efficacy and neurovascular regulatory mechanisms of Siguan points-based acupuncture treatment (SPBAT) for migraine.
Methods: A comprehensive literature search was conducted across 15 databases through January 1st, 2025.
Background: In recent years, emerging clinical research has prioritized assessment of combined therapeutic efficacy and safety parameters when programmed death 1 or its ligand (PD-1/L1) inhibitors are incorporated into first-line standard-of-care (SOC) therapy for metastatic colorectal cancer (mCRC). However, data obtained from these trials demonstrated conflicting evidence concerning survival benefits and clinical outcomes.
Aim: To evaluate the therapeutic impact and safety parameters of combining PD-1/L1 inhibitors with SOC protocols as first-line treatment for mCRC.
Unlabelled: Assessing lymph node metastasis (LNM) involvement in patients with rectal cancer (RC) is fundamental in disease management. In this study, we used artificial intelligence (AI) technology to develop a segmentation model that automatically segments the tumor core area and mesangial tissue from magnetic resonance T2-weighted imaging (T2WI) and apparent diffusion coefficient (ADC) images collected from 122 RC patients to improve the accuracy of LNM prediction, after which omics machine modeling was performed on the segmented ROI. An automatic segmentation model was developed using nn-UNet.

Thinning is a crucial silvicultural practice in forest management, the rational intensity of which plays an important role in increasing carbon sequestration capacity of forest ecosystems. However, it is not clear how different thinning intensities affect forest ecosystem carbon stocks and their fractions. We investigated plantations in the mountainous regions of eastern Liaoning Province, analyzed changes in carbon stocks and fractions with different thinning intensities (0, 10%-30%, 30%-50% and 50%-70%), and explored key factors influencing stand productivity and soil organic carbon dynamics.

To investigate the photosynthetic responses of needles with different ages (current-year and annual-year) in to sunflecks along the vertical gradient of canopy, we conducted an experiment at the Changbai Mountain Forest Ecosystem Positioning Station utilizing a canopy tower crane platform. We selected current-year and annual-year needles from the upper (mean height: 23.26 m), middle (16.

Despite being the most commonly mutated proteins in cancer, oncogenic RAS proteins remain largely untapped as pharmacological targets. Here, we report a synthetic cancer-killing platform, termed 'RAS-activated cancer killing (RACK)' system. Leveraging a transcriptional sensor designed to detect oncogenic RAS signals with high specificity, RACK achieves targeted identification and elimination of RAS-mutant cancer cells.

Methods: We followed the PRISMA guidelines to conduct a systematic review of articles from PubMed and Web of Science published between January 1, 2023 and November 9, 2024. Multiple reviewers including biomedical informaticians and a clinician involved in the article reviewing process.
: Elevated serum uric acid levels are associated with the occurrence, development, and adverse events of coronary heart disease (CHD) and CHD risk factors. However, the extent of any pathogenic effect of the serum uric acid on CHD and whether CHD risk factors play a confounding or mediating role are still unclear. : The potential causal associations of serum uric acid with CHD were evaluated via cross-trait linkage disequilibrium score regression analysis and Mendelian randomization.

Rationale: Glioblastoma is a fast-growing, invasive brain tumor with poor prognosis. Polyphyllin I, a saponin from Liliaceae plants, shows anti-tumor, anti-inflammatory effects, and induces ferroptosis, but its impact on glioblastoma and underlying mechanisms are not well understood.
Methods: Tumor malignancy was evaluated using assays such as scratch assays, CCK-8, clonogenic analyses, transwell experiments, along with EdU incorporation staining.
Alumina ceramic substrates are ideal materials for next-generation microelectronic systems and devices, widely used in aerospace, 5G communications, and LED lighting. High-quality hole processing is essential for system interconnection and device packaging. Millisecond lasers have emerged as a promising choice for hole processing in alumina ceramic due to their high processing efficiency.

In this paper, the electronic structure and photoelectric properties of graphdiyne nanotubes with armchair (A-GDYNT) and zigzag (Z-GDYNT) types have been studied. Calculations show that as n decreases, the divergence in gap values between (n)-A-GDYNT and (n)-Z-GDYNT increases. This is mainly attributed to the edge effect arising from their different boundaries.

Emerging evidence indicates that membraneless organelles stress granules (SGs), whose assemblies are driven by scaffold protein G3BP1, are extensively involved in tumor, especially in OS. However, how SGs behave and communicate with organelles, particularly nucleoli and mitochondria, during drug challenges remain unknown.

Background: Diagnosis of venous thromboembolism (VTE) is often delayed, and facilitating earlier diagnosis may improve associated morbidity and mortality. Clinical notes contain information not found elsewhere in the medical record that could facilitate timely VTE diagnosis and accurate quality measurement. However, extracting relevant information from unstructured clinical notes is complex.

Background: Parkinson's disease (PD) is a neurodegenerative disorder with some progressive impairment and an unclear pathogenesis.
Purpose: This study aimed to use resting-state functional magnetic resonance imaging (rs-fMRI) and graph analysis approaches to compare changes in brain functional network topology in PD at different disease stages.
Materials And Methods: A total of 58 PD patients, comprising 29 early-stage PD (PD-E) and 29 middle-to-late stage PD (PD-M), and 29 age- and sex-matched healthy control (HC) participants, were recruited.
The bioprocess of recycling polyethylene terephthalate (PET) has been extensively studied; however, challenges remain due to its process complexity, low enzymatic performance, and high costs. Here, we devised a strategy termed the Integrated Self-Assembling Multi-Enzyme Display Platform (ISA-MEDP) to directly and efficiently degrade postconsumer PET (pc-PET). We developed a novel surface display system for the unconventional and robust yeast Candida tropicalis, enabling FAST-PETase and MHETase to be functionally anchored onto the cell wall.

Increasing species richness is believed to mitigate the effects of metal contamination, thereby enhancing phytoremediation. However, the overall effects of increased species richness on plant remediation under Cd contamination, along with the underlying mechanisms, remain unclear.

Large yellow croaker (Larimichthys crocea) is one of the most important marine economic fish. However, due to overfishing and long-term artificial breeding, the germplasm has degraded in recent years, and the wild germplasm has been listed as critically endangered. It is urgent to protect the germplasm resources of Larimichthys crocea.

Conventional syntheses of robust Zr-based metal-organic frameworks (Zr-MOFs) rely on harsh solvothermal conditions, precluding the inclusion of fragile functionalities such as enzymes or organisms. Therefore, developing routes to such stable frameworks under ambient conditions remains a significant challenge. Here, we report a mild aqueous solid-state crystallization (SSC) strategy that enables Zr-MOF assembly at ambient temperature.
