Despite advancements in systemic therapy, the mortality rate for patients with metastatic melanoma remains around 70%, underscoring the imperative for alternative treatment strategies. Through the establishment of a chemoresistant melanoma model and a subsequent drug investigation, we have identified pacritinib, a medication designed for treating myelofibrosis and severe thrombocytopenia, as a potential candidate to overcome resistance to melanoma therapy. Our research reveals that pacritinib, administered at clinically achievable concentrations, effectively targets dacarbazine-resistant melanoma cells by suppressing IRAK1 rather than JAK2.

Different starch crystal structures significantly influence meat product quality, though their specific impacts on myofibrillar protein (MP) functionality remain unclear despite industry demand for optimized ingredients. This study compared how potato, corn, mung bean, and pea starches affect MP properties in minced pork. Our findings reveal that starch-protein interactions fundamentally regulate MP gel and emulsion properties through the following mechanisms: First, starch promotes protein aggregation by enhancing hydrophobic interactions and disulfide bond formation, affecting gel network crosslinking.

The optimized NiFe-PyThr-4:1 catalyst exhibits remarkable OER activity, requiring low overpotentials of only 162 and 222 mV to achieve current densities of 10 and 100 mA cm, respectively, along with a small Tafel slope of 34.

We introduce a cell-free DNA (cfDNA) fragmentation pattern: the fragment dispersity index (FDI), which integrates information on the distribution of cfDNA fragment ends with the variation in fragment coverage, enabling precise characterization of chromatin accessibility in specific regions. The FDI shows a strong correlation with chromatin accessibility and gene expression, and regions with high FDI are enriched in active regulatory elements. Using whole-genome cfDNA data from five datasets, we developed and validated the FDI-oncology model, which demonstrates robust performance in early cancer diagnosis, subtyping, and prognosis.
