Heart failure (HF) and lung cancer (LC) often coexist, yet their shared molecular mechanisms are unclear. We analyzed transcriptome data from the NCBI Gene Expression Omnibus (GEO) database (GSE141910, GSE57338) to identify 346 HF‑related differentially expressed genes (DEGs), then combined weighted gene co-expression network analysis (WGCNA) pinpointed 70 hub candidates. Further screening of these 70 hub candidates in TCGA lung cancer cohorts via LASSO, Random Forest, and multivariate Cox regression suggested CYP4B1 as the only independent prognostic marker.

Background: Lactate has been shown to play an important immunosuppressive role in the tumor microenvironment (TME) and promote tumor progression through a variety of different mechanisms of action. Myeloid-derived suppressor cells (MDSCs) are important cells that play an immunosuppressive role in the TME. However, the underlying mechanism by which lactate regulates MDSCs remains unclear.

Protein phosphorylation modification plays an important role in regulating protein activity. Astrocyte elevated gene-1 (AEG-1), an adaptor protein, promotes the progression of various types of cancers by protein-protein interactions. We previously demonstrated that AEG-1 promoted the growth and metastasis of gastric cancer by upregulating the expression of oncogenic eukaryotic translation initiation factor 4E (eIF4E).

Endometrial cancer (EC) presents a major global health challenge due to its heterogeneity and complex pathophysiology. The tumor microenvironment (TME), comprising stromal cells, immune cells, endothelial cells, and non-cellular components, critically influences EC progression. Single-cell RNA sequencing (scRNA-seq) has revolutionized our understanding of EC by revealing TME cellular heterogeneity and interactions.

ARPC1B cancer stem cells (CSCs) in pancreatic cancer are identified as a subpopulation resistant to gemcitabine. In our study, drug repositioning, molecular docking, and surface plasmon resonance (SPR) technique jointly revealed that CK-636 can directly target ARPC1B protein with high affinity. In vitro cytotoxicity, ex vivo organoid cultures, in vivo xenograft and orthotopic gemcitabine-resistant pancreatic cancer model demonstrated that combination therapy of gemcitabine plus CK-636 showed a superior anti-tumor effect compared with gemcitabine monotherapy.

Tenuifolin Attenuates Methamphetamine-Induced Reinstatement in Mice by Regulating Hippocampal Postsynaptic BDNF Signaling.

CNS Neurosci Ther

August 2025

Department of Substance Dependence, The Affiliated Xuzhou Eastern Hospital of Xuzhou Medical University, Xuzhou Eastern People's Hospital, Xuzhou, China.

Background: Compulsive relapse (reinstatement) behavior of methamphetamine underlies the difficulty of withdrawal and is associated with abnormal BDNF-mediated synaptic plasticity. However, how to intervene in this aberrant synaptic plasticity to prevent its reinstatement behavior in mice has not fully been studied.

Methods: The CPP was used to establish a model of methamphetamine-induced reinstatement behavior in C57BL/6 mice.

Background: Extracellular matrix-related genes (ERGs) are crucial in the tumorigenesis of various malignancies, including cervical cancer (CC), but their prognostic significance in CC has not been thoroughly investigated.

Methods: We interrogated RNA-seq expression profiles from the public datasets to identify differentially expressed ERGs. Cox regression analysis was utilized to evaluate their prognostic significance.

The limited response rate to immune checkpoint inhibitors (ICIs) remains a significant challenge in the treatment of lung adenocarcinoma (LUAD). In our study, we identified a lactate-based chemical barrier surrounding FAP cancer-associated fibroblasts (CAFs) within the LUAD microenvironment (TME), which may hinder the infiltration and function of CD8 T cells. Further investigation revealed that FAP CAFs specifically overexpress LINC01711, which drives lactate production by promoting FGFR1-mediated phosphorylation of lactic dehydrogenase A (LDHA) at the Y10 site and facilitating the formation of active LDHA tetramers.

Background: Lung cancer, a leading cause of cancer death, displays profound histologic and molecular heterogeneity across adenocarcinoma, squamous, and small-cell types. Clinically, tumours can shift between these states, reflecting lineage plasticity-the reprogramming of differentiated cells to alternate identities. Pre-existing genomic/epigenomic diversity and microenvironmental cues supply the substrates and pressures for plasticity from disease onset.

Liquid-Liquid Phase Separation (LLPS) refers to the separation of biomacromolecules into separate liquid phases within the cells, plays a critical role in lung cancer pathogenesis. Using machine learning, we developed an LLPS-associated signature (LLPSAS) based on 79 key genes. The LLPSAS demonstrated superior prognostic performance compared to 140 existing lung adenocarcinoma (LUAD) prognostic models.

Globally, breast cancer remains one of the most prevalent malignancies and a leading cause of cancer-related death in women, with over 2.3 million new cases reported annually. Despite treatment advances, one breast cancer type in particular, HER2-negative breast cancer, lacks precise therapeutic targets.

Colorectal cancer (CRC) is a leading cause of cancer-related mortality worldwide. Epidemiological studies suggest that dietary fiber intake reduces CRC risk, with butyrate, a fermentation product of dietary fiber, showing promising anti-cancer properties. However, the underlying molecular mechanisms remain poorly understood.

The role of meningeal lymphatic vessels (mLVs) in neurodegenerative diseases has been increasingly recognized. However, their involvement in lipopolysaccharide (LPS)-induced neuroinflammation and associated depression-like behaviors remains poorly understood. Given that impaired clearance of neurotoxic substances can prolong central nervous system (CNS) inflammation, investigating the function of mLVs in this context may offer new insights into the mechanisms underlying acute neuroinflammation and provide potential therapeutic targets.
