Hidroacanthoma simplex (HS) is a rare skin appendage tumor that typically appears on the trunk and lower limbs in the elderly. Although HS is a predominantly benign condition, the presence of cellular atypia and dermal infiltration on histological examination indicates malignant HS (MHS). 5-aminolaevulinic acid photodynamic therapy (ALA-PDT) uses a photosensitizer and corresponding light source to cause irreversible damage or death of target cells through a photochemical reaction.

In clinical practice, tumor-targeting diagnosis and immunotherapy against programmed death ligand 1 (PD-L1) have a significant impact. In this research, a PD-L1-antagonistic affibody dimer (Z) was successfully prepared through Escherichia coli expression system, and conjugated with the photosensitizer of ICG via N-hydroxysuccinimide (NHS) ester to develop a novel tumor-targeting agent (ICG-Z) for both tumor imaging diagnosis and photothermal-immunotherapy simultaneously. In vitro, Z could specifically bind to PD-L1-positive LLC and MC38 tumor cells, and ICG-Z-mediated photothermal therapy (PTT) also showed excellent phototoxicity to these tumor cells.

Xeroderma pigmentosum(XP) is a rare autosomal recessive genodermatosis. Individuals with XP are characterized by severe skin sensitivity to sunlight, and more susceptible to the development of skin malignancies in sun-exposed regions. 5-Aminolevulinic acid photodynamic therapy (ALA-PDT) is an effective modality for vulvar lichen sclerosus (VLS) and squamous cell carcinoma (SCC) in situ. Here we first describe a case of VLS that relapsed after ALA-PDT without follow-up and progressed to SCC eventually. The first two courses of ALA-PDT (total of 21 sessions) were effective for VLS.

Background: Extramammary Paget's Disease (EMPD) is an intraepithelial cancer that is prone to recurrence and sometimes refractory to therapy. A few EMPD cases have been treated with Photodynamic therapy (PDT), which reported high complete remission (CR) rates and low recurrence with hematoporphyrin derivatives (HpD) The aim of this study was to further explore the efficacy and safety of HpD-PDT for EMPD patients.
Methods: Open-label, single arm, pilot study was designed to investigate the role of HpD-PDT in EMPD.
Herein, alkenylpiperidine and alkynylpiperidine moieties were introduced into the left wing of DAPYs (diarylpyrimidines) to explore the new site of the NNIBP (non-nucleoside inhibitor binding pocket) protein-solvent interface region via the structure-based drug design strategy.
