R-loop, a three-stranded nucleic acid structure consisting of the RNA:DNA hybrid and the displaced singlestranded DNA, is crucial for many cellular processes but could be a threat to genome integrity if dysregulated. The homeostasis of R-loops is governed by various factors including helicases, nucleases, and chromatin remodelers. Since there are many excellent reviews about R-loops, we focus on discussing how R-loop homeostasis is regulated via nucleic acid and protein modifications.

Article Synopsis
  • Cytomegalovirus reactivation (CMVr) and bloodstream infections (BSI) are significant complications for patients after allogeneic hematopoietic stem cell transplantation (allo-HSCT), leading to high morbidity and mortality rates.
  • A study involving 500 CMV DNA-negative and IgG-positive allo-HSCT recipients found that 400 had CMVr and 75 experienced BSI within the first 100 days post-transplant, showcasing graft failure and acute graft-versus-host disease as key risk factors affecting prognosis.
  • The research highlights that while BSI increases short-term mortality risk, surviving BSI can lower long-term risk; however, the combination of CMVr and BSI severely impacts survival outcomes in patients.

RNF214 is an understudied ubiquitin ligase with little knowledge of its biological functions or protein substrates. Here we show that the TEAD transcription factors in the Hippo pathway are substrates of RNF214. RNF214 induces non-proteolytic ubiquitylation at a conserved lysine residue of TEADs, enhances interactions between TEADs and YAP, and promotes transactivation of the downstream genes of the Hippo signaling.

Self-supervised Object Segmentation (SOS) aims to segment objects without any annotations. Under conditions of multi-camera inputs, the structural, textural and geometrical consistency among each view can be leveraged to achieve fine-grained object segmentation. To make better use of the above information, we propose Surface representation based Self-supervised Object Segmentation (Surface-SOS), a new framework to segment objects for each view by 3D surface representation from multi-view images of a scene.

Histone H2A monoubiquitination (H2Aub1) functions as a conserved posttranslational modification in eukaryotes to maintain gene expression and guarantee cellular identity. Arabidopsis H2Aub1 is catalyzed by the core components AtRING1s and AtBMI1s of polycomb repressive complex 1 (PRC1). Because PRC1 components lack known DNA binding domains, it is unclear how H2Aub1 is established at specific genomic locations.
