Abnormal tumor homeostasis is crucial in maintaining tumor cell survival and growth. Thus, developing effective antitumor strategies that can simultaneously disrupt multiple intracellular homeostatic mechanisms may be more effective and less laborious. Herein, an autoamplificatory nanoinducer (CLRN) composed of photosensitizer Ce6, glucose metabolism inhibitor lonidamine and methyltransferase inhibitor RG108 is designed and constructed to enhance pyroptosis and upregulate immunosurveillance by disrupting intracellular redox and metabolic homeostasis.

Prior research has yielded inconsistent results regarding the relationships between individual essential metal elements (EMEs) and gallstone disease (GSD). Furthermore, the association between the EME mixture and GSD, as well as the underlying mechanisms, remains poorly understood. We aimed to assess the associations between individual EMEs and their mixture with GSD, and to investigate the mediation roles of superoxide dismutase (SOD).

Introduction: Multiple studies have shown that genetic polymorphism in the MTHFR gene is associated with susceptibility to type 2 diabetes mellitus (T2DM), but the results remain controversial. This study was divided into two parts. The first part was to explore the relationship between the SNPs of MTHFR gene (C677T and A1298C) and genetic susceptibility to T2DM.

Background: The combined effects of physical activity (PA) and nitrogen dioxide (NO) on fasting plasma glucose (FPG) levels have rarely been studied. This study aimed to examine whether long-term exposure to NO attenuates the association between PA and FPG levels in non-diabetic older adults.

Methods: A total of 2600 non-diabetic elderly Chinese adults were included in this cross-sectional study.

RelA, also known as nuclear factor kappa B p65, plays a crucial role in the pathogenesis of various liver diseases. However, the specific role of RelA in hepatocytes during the progression of metabolic dysfunction-associated steatotic liver disease (MASLD) is not well understood. This study explored the relationship between impaired RelA signaling and lipid metabolism disorders in hepatocytes, and how they synergistically contribute to the advancement of MASLD.

Ferroptosis combined with photodynamic therapy (PDT) has emerged as a powerful approach to induce cancer cell death by producing and accumulating lethal reactive oxygen species (ROS) in the tumor microenvironment (TME). Despite its efficacy and safety, challenges persist in delivering multiple drugs to the tumor site for enhanced antitumor efficacy and improved tissue targeting. Hence, we designed a method of inducing ferroptosis through laser-mediated and human homologation-specific efficient activation, which is also a ferroptosis therapy with higher safety through ROS-mediated.

The gut microbiome primarily generates short-chain fatty acids (SCFAs) by fermenting dietary fibers. Though previous studies have linked SCFAs to blood pressure, there remains a lack of research on the relationship between SCFAs levels in the serum of elderly individuals and blood pressure. Based on this, we investigated the associations of serum SCFAs with blood pressure in Chinese older adults in a cross-sectional study.

There is limited research on the effects of non-essential metal (NEM) mixture on handgrip strength in the elderly. This study aimed to assess the associations of single NEMs and their mixture with handgrip strength in Chinese community-dwelling older adults. A total of 3807 elderly people aged 60 years or above were included in this study.

Article Synopsis
  • - The study aimed to assess the links between seven metal(loid)s (lead, manganese, nickel, arsenic, cadmium, chromium, vanadium) and hypertension in older Chinese adults, involving 1009 participants whose blood metal(loid) levels were measured.
  • - Findings showed a significant association between lead (Pb) and hypertension, with logistic regression revealing an odds ratio indicating higher hypertension risk among those with elevated Pb levels; this association remained when considering multiple metal(loid)s together.
  • - The research highlighted that the relationship between the metal(loid) mixture and hypertension could be influenced by factors like diabetes, age, BMI, and area, with Pb being the most significant metal affecting hypertension in older adults.

Abnormal tumor metabolism creates a complex tumor immune microenvironment that plays a dominant role in the metastasis of triple-negative breast cancer (TNBC). TNBC is insensitive to immune checkpoint blockade (ICB) therapy because of insufficient cytotoxic T lymphocyte (CTL) infiltration and a hyper-lactic acid-suppressive immune microenvironment caused by abnormal glycolysis. Herein, we propose an amplified strategy based on lactic acid regulation to reprogram the immunosuppressive tumor microenvironment (ITM) and combine it with ICB therapy to achieve enhanced antitumor immunotherapy effects.

Sleep quality significantly affects the quality of life of older persons. Therefore, this study explored the relationship between sleep quality and living environment of older persons in China to provide a theoretical basis for therapies to alleviate sleep disorders in older persons. A total of 6211 subjects > 60 years of age in Anhui Province, China, were evaluated using the Pittsburgh Sleep Quality Index and a self-reported questionnaire.

Background: Job burnout is a prevalent and emerging challenge in the primary medical system, causing mass turnover, especially of primary medical staff. Little attention has been paid to the different dimensions of job burnout (emotional exhaustion, personality disintegration, and reduced sense of achievement), which may hinder efforts to tackle high turnover intention among primary medical staff. From the perspective of conservation of resources theory, social support and psychological capital are basic resources with potential to diminish job burnout and thus lower turnover intention.

Previous studies have related single toxic metals (TMs) to hyperuricemia (HUA) among the general population, however, the association of the TM mixture with HUA, especially in older adults, remains poorly understood. We aimed to examine the relationships between individual TMs and their mixture and HUA in Chinese rural older adults. This study consisted of 2075 rural older adults aged 60 years or over.
