Unlabelled: Yarrowia lipolytica is widely used for the industrial production of the natural sweetener erythritol. Despite improvements in fermentation process control and metabolic pathway regulation, bottlenecks still exist in terms of yield and screening technology. Therefore, we constructed an artificial sensor system for effective erythritol detection, established a single-cell droplet-based high-throughput screening system based on fluorescence-activated cell sorting, and obtained Y. lipolytica with improved erythritol production through mutagenesis and high-throughput screening. We used a droplet generator to co-cultivate Y. lipolytica 5-14 with Escherichia coli and used the E. coli fluorescent signal to detect the concentration of erythritol synthesized by Y. lipolytica 5-14 for high-throughput screening. Strains were subjected to UV mutagenesis for 120 s. Under optimized fermentation conditions using Y. lipolytica mutants in 96-well plates, the screening efficiency reached 16.7%. Yarrowia lipolytica 5-14-E6 showed a 21% increase in erythritol to 109.84 g/L. After fermentation at 30°C in a 100-m3 fermenter for 75 hr, the mutant Y. lipolytica 5-14-E6 erythritol yield reached 178 g/L.
One-sentence Summary: In this study, we constructed an artificial sensor system for effective erythritol detection, established a single-cell droplet-based high-throughput screening system based on fluorescence-activated cell sorting, and induced and screened mutant strains of Yarrowia lipolytica with improved erythritol production through mutagenesis and high-throughput screening.

Mycoplasma gallisepticum (MG) is one of the main pathogens causing chronic respiratory diseases in chickens, which seriously affects the sustainable and healthy development of the poultry industry and leading to heavy economic losses. Therefore, we developed a safe, efficient, convenient, and low-cost MG oral vaccine. The vaccine is based on a recombinant yeast surface display system to compensate for the shortcomings of existing vaccines.

The function of fermented and non fermented bamboo-derived feed additives in poultry nutrition is critically assessed in this review, with emphasis on the effects on growth performance, immunity, intestinal health, egg and meat quality. Fermented bamboo feeds have become a promising nutritional innovation in poultry production. The use of bamboo leaves and tender shoots in chicken feed is limited due to the presence of lignin and cellulose.

The Fabaceae-specific review highlights the structural, functional, and phylogenetic diversity of UGTs, revealing clade-specific glycosylation mechanisms and novel sugar conjugations that contribute to legume adaptability. These insights offer promising avenues for metabolic engineering and stress-resilient crop development. UDP-glycosyltransferases (UGTs) are the biocatalysts modifying small molecules through glycosylation to enhance their solubility, stability, and bioactivity.

High-throughput screening (HTS) assays are pivotal in modern biomedical research, particularly in drug discovery and functional genomics. Ensuring the quality and reliability of HTS data is critical, especially when dealing with the small sample sizes that are typical in such assays. This study explores the integration of two powerful statistical metrics-Strictly Standardized Mean Difference (SSMD) and Area Under the Receiver Operating Characteristic Curve (AUROC)-for quality control (QC) in HTS.

The auditory brainstem response (ABR) remains the gold standard for evaluating hearing function in both animal models and humans. Features of ABR, including threshold, wave I amplitude and latency are critical for diagnosing and investigating the mechanisms of hearing loss. Critically, the rapid proliferation of genetically engineered mouse models in hearing research has created an imperative demand for high-throughput ABR testing capabilities.
