Taiwania cryptomerioides Hayata is an endangered relict plant belonging to Taxodiaceae, and it is also an endemic plant to China. The decay-resistant of Taiwania timber can provide highly quality wood for building and furniture. Plenty of regenerative of leaves of T. cryptomerioides also has been used as a resource for the discovery of new dimeric diterpenoids. In a search for structurally diverse dimeric diterpenoids and potent bioactive isolates, ten new heterodimeric diterpenoids, taiwaniadducts K-T (1-4, 6, 8-11, and 14), along with five known ones (5, 7, 12, 13, and 15), were isolated from the leaves of T. cryptomerioides. These new compounds were defined by comprehensive spectroscopic analyses, putative biosynthetic pathways, and the values of optical. Biologically, anti-multidrug resistance (MDR) activities of compounds were evaluated. Compounds 4 and 10 exerted a 9.18-fold potentiation effect on bortezmib (BTZ) susceptibility at a tested concentration (20 μM) better than the positive control verapamil. The research of the leaves of T. cryptomerioides not only added the new data to the structural diversity and activities of dimeric diterpenoids but also could provide support for the medical and industrial application of the leaves of this endangered relict plant.

Disrupting redox balance by elevating reactive oxygen species (ROS) selectively induces cancer cell death while sparing healthy tissues. Peptidyl-prolyl isomerase (Pin1) is overexpressed in several cancer cells and plays a critical role in tumorigenesis and tumor progression. All-trans retinoic acid (RA) is known to induce cell death by generating ROS and directly binding the active site of Pin1, leading to cancer cell death.

The intratracheal route enables an enhanced drug accumulation within the lungs together with reduced systemic exposure, making it a promising approach for treating epidermal growth factor receptor (EGFR)-overexpressed non-small cell lung cancer (NSCLC). In this study, dextran-PEG-erlotinib co-polymers (DPE), novel amphiphilic conjugates with outstanding therapeutic activities against EGFR-overexpressed NSCLC and stabilizing effects were synthesized and facilitated self-assembly of redox-sensitive paclitaxel dimers. The obtained nanoassemblies (DPE dimer NPs) exhibited high drug loading efficiency, satisfactory stability and optimal redox responsive paclitaxel release profile comparable with control nanoassemblies (TPGS dimer NPs), where paclitaxel dimers were assembled in the presence of TPGS, a therapeutically inert stabilizer.

This study reports a novel case of acute promyelocytic leukemia (APL) characterized by a tripartite fusion gene, NUP98::RARG::LINE-L2a, formed by the rearrangement of nucleoporin 98 (NUP98), retinoic acid receptor gamma (RARG), and long interspersed nuclear element L2a (LINE-L2a). The molecular mechanism underlying the patient's resistance to all-trans retinoic acid (ATRA) is also investigated. The 32-year-old male was admitted to Shandong Provincial Hospital Affiliated to Shandong First Medical University with complaints of "a 10-day cough and newly detected leukocytosis for one day".

We report the design of folded cyclodipeptides that show dynamic and adaptive conformational transitions responding to solvents and enantioselective recognition of a biomolecule, vitamin K1 (VK1). Postmodification of phenylalanine-based cyclodipeptides with polycyclic aromatic hydrocarbons (pyrene, anthracene, and phenanthrene) produced chiral folded structures. Compared with other solvents, these cyclodipeptides exhibit ultrahigh sensitivity to dimethyl sulfoxide (DMSO), which induces a conformational transition from parallel to perpendicular dimerization.
