Cadmium (Cd) pollution in soils poses a serious threat to crop production and human health. Although nitric oxide (NO) is known to mitigate metal stress, its role in response to Cd stress remains controversial. This study investigated the underlying mechanisms by which NO mitigates Cd toxicity in soybean.

Cytosolic G6PDH contributes to soybean tolerance to heavy metal exposure by alleviating oxidative damage, and in this process, nitric oxide may act upstream of cytosolic G6PDH. Heavy metal (HM) pollution in soil significantly impairs agricultural production and represents a substantial risk to food security. However, the underlying mechanism by which glucose-6-phosphate dehydrogenase (G6PDH or G6PD, EC 1.

Neuroblastoma, the most prevalent extracranial pediatric solid tumor, arises from neural crest progeny cells. It exhibits substantial developmental plasticity and intratumoral heterogeneity, leading to survival rates below 50% in high-risk cases. The regulatory mechanisms underlying this plasticity remain largely elusive.

Background: Glutaredoxins (GRX) are key oxidoreductases that modulate protein redox states during plant development and stress responses. Alkaligrass (Puccinellia tenuiflora) is a highly salt-tolerant forage grass, but its GRX gene family (PutGRXs) remains uncharacterized, unlike those in Arabidopsis and other plants.

Results: We identified 25 PutGRX genes in the P.

T-cell acute lymphoblastic leukemia (T-ALL) is an aggressive leukemia subtype and a prevalent malignancy in children, with poor prognosis, high relapse rates, and drug resistance. Recent research has shown that super-enhancer-regulated genes play crucial roles in T-ALL progression. In this study, we identified SH2 domain containing 1 A (SH2D1A) as a gene regulated by super-enhancers, and is overexpressed, which correlates with unfavorable clinical outcomes in T-ALL.

Background: Acute megakaryoblastic leukemia (AMKL) is a rare hematological malignancy in adults but children. Alternative splicing (AS) has been shown to affect hematological cancer progression, making splicing factors promising targets. Our research aims to investigate the efficacy of the molecular glue degrader indisulam, which targets the splicing factor RNA binding motif protein 39 (RBM39) in AMKL models.

Dysregulated amino acid metabolism is a key contributor to polycystic ovary syndrome (PCOS). This cross-sectional study revealed that serum levels of L-kynurenine (L-Kyn) were significantly elevated in women with PCOS, whereas pyridoxal 5'-phosphate (PLP) levels were markedly reduced. Moreover, human serum L-Kyn levels exhibited a positive correlated with low-density lipoprotein cholesterol (LDL-C) and a negative correlation with high-density lipoprotein cholesterol (HDL-C).

Cell cycle dysregulation and the corresponding metabolic reprogramming play significant roles in tumor development and progression. CDK9, a kinase that regulates gene transcription and cell cycle, also induces oncogene transcription and abnormal cell cycle in AML cells. The function of CDK9 for gene regulation in AML cells requires further exploration.

Background: Chronic salpingitis is one of the most common causes of female infertility. Luteal support is a critical step for embryo transfer. Here, we evaluated the effects of two luteal support regimens, intramuscular progesterone (IMP) and progesterone vaginal gel (VAG), on the pregnancy outcomes in patients with chronic salpingitis undergoing vitrified-warmed embryo transfer.

Article Synopsis
  • Super enhancers (SEs) are essential for cell identity and can activate oncogenes in cancer, particularly in T-cell acute lymphoblastic leukemia (T-ALL).
  • A new SE has been found in T-ALL that regulates the gene IRF2BP2, influenced by key transcription factors like ERG and ELF1.
  • While IRF2BP2 does not significantly affect normal T cell development, it is vital for T-ALL cell growth and survival, implicating it as a potential target for treatment options.

Article Synopsis
  • Pediatric T-cell acute lymphoblastic leukemia (T-ALL) still poses significant challenges, including treatment resistance and recurrence, despite advances in prognosis, with LIM domain-binding protein 1 (LDB1) playing a crucial role in this context.
  • * Researchers utilized techniques like shRNA, RNA-Seq, and CUT&Tag to analyze LDB1's function in T-ALL cell lines and uncovered its cooperation with key transcription factors (ERG, ETV6, IRF1) to regulate downstream targets.
  • * The study concludes that LDB1 is essential for T-ALL cell proliferation and survival, specifically regulating the significant target gene MYB, thus providing insights into its role in T-ALL progression.*

Background: Osteosarcoma (OS) is one of the most common primary malignant tumors of bone in children, which develops from osteoblasts and typically occurs during the rapid growth phase of the bone. Recently, Super-Enhancers(SEs)have been reported to play a crucial role in osteosarcoma growth and metastasis. Therefore, there is an urgent need to identify specific targeted inhibitors of SEs to assist clinical therapy.

Background: Despite the use of targeted therapeutic approaches, T-cell acute lymphoblastic leukemia (T-ALL) is still associated with a high incidence of complications and a poor prognosis. Indisulam (also known as E7070), a newly identified molecular glue compound, has demonstrated increased therapeutic efficacy in several types of cancer through the rapid degradation of RBM39. This study aimed to evaluate the therapeutic potential of indisulam in T-ALL, elucidate its underlying mechanisms and explore the role of the RBM39 gene.

Background: Super-enhancers (SEs) typically govern the expression of critical oncogenes and play a fundamental role in the initiation and progression of cancer. Focusing on genes that are abnormally regulated by SE in cancer may be a new strategy for understanding pathogenesis. In the context of this investigation, we have identified a previously unreported SE-driven gene IRF2BP2 in neuroblastoma (NB).

Neuroblastoma (NB) is a common childhood tumor with a high incidence worldwide. The regulatory role of RNA N6-methyladenosine (m6A) in gene expression has attracted significant attention, and the impact of methyltransferase-like 14 (METTL14) on tumor progression has been extensively studied in various types of cancer. However, the specific influence of METTL14 on NB remains unexplored.

Super enhancers (SEs) are genomic regions comprising multiple closely spaced enhancers, typically occupied by a high density of cell-type-specific master transcription factors (TFs) and frequently enriched in key oncogenes in various tumors, including neuroblastoma (NB), one of the most prevalent malignant solid tumors in children originating from the neural crest. Cyclin-dependent kinase 5 regulatory subunit-associated protein 3 (CDK5RAP3) is a newly identified super-enhancer-driven gene regulated by master TFs in NB; however, its function in NB remains unclear. Through an integrated study of publicly available datasets and microarrays, we observed a significantly elevated CDK5RAP3 expression level in NB, associated with poor patient prognosis.

ANP32B, an important nuclear protein associated with proliferation, has been found to regulate hematopoiesis and CML leukemogenesis by inhibiting p53 activity. However, recent study suggests that ANP32B exerts a suppressive effect on B-cell acute lymphoblastic leukemia (ALL) in mice by activating PU.
