Article Abstract

Background: Papillary thyroid carcinoma (PTC) is a globally widespread inflammation-related cancer, where lymph node metastasis (LNM) poses significant challenges to the prognosis of PTC. The role of the monocyte-to-high-density lipoprotein cholesterol ratio (MHR), a novel inflammation marker attracting increasing attention, in PTC remains unclear.

Methods: Clinical data analysis was adopted to preliminarily explore the relationship between clinical features and LNM of PTC. Single-cell RNA sequencing (scRNA-seq) data from the GSE191288 and GSE193581 datasets were integrated to analyze various single-cell infiltration in PTC. A comprehensive suite of in vitro experiments, encompassing immunohistochemistry, co-culture, and Transwell assays were conducted to elucidate the regulatory role of macrophages and PTC.

Results: Clinical data analysis confirmed MHR could be an independent risk factor for LNM (OR = 1.76, 95%CI: 1.20-2.88, P = 0.011). The single-cell analysis identified cell clusters associated with dysregulated cholesterol homeostasis Q1 and CD68 + C3 macrophage subpopulations, as well as their markers GDF15. Further analysis confirmed that they are closely related to PTC metastasis and malignancy, which also implied a significant correlation between MHR and LNM. The in vitro experiments demonstrated PTC may promote metastasis by mediating inducing macrophage differentiation to the M2 phenotype.

Conclusion: This study revealed the potential role of MHR in PTC from the single-cell perspective for the first time. Combined with the results of clinical studies and basic experiments, it was confirmed that mononuclear/macrophage and cholesterol homeostasis significantly promoted the lymph node metastasis of PTC. Overall, these findings informed robust support for MHR as an emerging marker for preoperative LNM prediction of PTC.

Objectives: Increasing detection of low-risk papillary thyroid carcinoma (PTC) is associated with overdiagnosis and overtreatment. N6-methyladenosine (mA)-mediated microRNA (miRNA) dysregulation plays a critical role in tumor metastasis and progression. However, the functional role of mA-miRNAs in PTC remains unclear.

Second Primary Papillary Thyroid Carcinoma: Insights From Competing Risk Analysis and Post-RAIT.

Division of Thyroid Surgery, Department of General Surgery, West China Hospital, Sichuan University, Chengdu, China.

Background: Improved cancer survival rates have highlighted second primary malignancies (SPMs), with the thyroid gland being one of the most common organs developing SPMs in cancer survivors. Second primary papillary thyroid carcinoma (2-PTC) is the predominant type, yet it remains poorly understood. This study aims to delineate the clinicopathological features and survival outcomes of 2-PTC and assess the efficacy of postoperative radioactive iodine therapy (post-RAIT) in reducing mortality risks in intermediate-risk 2-PTC patients.

Hypomyelinating leukodystrophy: From molecular mechanisms to clinical advances.

Hypomyelinating leukodystrophies (HLDs) are a group of inherited disorders characterized by impaired myelin formation in the central nervous system. Among them, Pelizaeus-Merzbacher disease (PMD) is a well-defined X-linked leukodystrophy caused by mutations in the PLP1 gene, including duplications, missense variants, and null mutations. Recent studies have revealed that different types of PLP1 mutations lead to distinct pathomechanisms: while missense mutations induce endoplasmic reticulum stress and activate the unfolded protein response (UPR), PLP1 duplications cause aberrant intracellular trafficking and cholesterol accumulation without UPR activation.

Downsizing Pt particles and incorporating water dissociation site represents a promising strategy for maximizing atomic utilization efficiency and enhancing catalytic performance in Pt-based hydrogen evolution reaction (HER) electrocatalysts. Here, we present a self-supported Pt/Y(OH) electrocatalyst through a synergistic combination of anion insertion-enhanced electrodeposition and chemical deposition at ambient temperature. The resultant architecture features sub-2 nm Pt nanoclusters (with an average diameter of 1.
