In recent years, the epigenetic regulation in tumor development has garnered significant attention. The lysine-specific demethylase 1 (LSD1) and histone deacetylase (HDAC), key enzymes involved in epigenetic modification, are overexpressed in various cancers and promote tumor cell proliferation and differentiation by modulating histone modifications. Although single-target inhibitors for LSD1 or HDAC exhibit efficacy in preclinical models, their clinical utility is hindered by compensatory signaling pathways and the emergence of drug resistance.

Osteoclasts, which are derived from myeloid precursors, are essential for physiologic bone remodeling but also mediate pathological bone loss in inflammatory diseases such as periodontitis and rheumatoid arthritis. Lysine-specific demethylase (LSD1/KDM1A) is a histone demethylase that modulates the chromatin landscape via demethylation of H3K4me1/2 and H3K9me1/2, thereby regulating the expression of genes essential for deciding cell fate. We previously demonstrated that myeloid-specific deletion of LSD1 (LSD1LysM-Cre) disrupts osteoclast differentiation, leading to enhanced BV/TV under physiological conditions.

Background: Immunotherapy combined with chemotherapy has become the standard treatment for HER2-negative gastric cancer (GC), but its clinical benefits remain limited, with a median progression-free survival (mPFS) of 6-8 months and median overall survival (mOS) of 15-18 months. These outcomes are particularly poor in patients with CPS < 1. The marked heterogeneity of GC, along with primary and secondary resistance, presents significant clinical challenges and underscores the urgent need for novel therapeutic strategies.
