Supersaturation and precipitation within the gastrointestinal tract can influence oral absorption of active pharmaceutical ingredients (APIs). Supersaturation of weakly basic APIs upon transfer from the stomach into the small intestine may enhance their absorption, while salt forms of poorly soluble weak acids may generate supersaturated solutions in both stomach and intestine. Likewise, APIs with solubility-limited absorption may be developed as enabling formulations intended to produce supersaturated solutions of the API in the gut. Integrating the supersaturation/precipitation characteristics of the API into the biopharmaceutical risk classification enables comprehensive mapping of potential developability risks and guides formulation selection towards optimizing oral bioavailability (BA). The refined Developability Classification System (rDCS) provides an approach for this purpose. In this work, the rDCS strategy is revisited and a stratified approach integrating the in vitro supersaturation and precipitation behavior of APIs and their formulations is proposed.

Crystalluria reflects the urinary physicochemical environment and may serve as a recurrence marker, but key crystals like brushite are rarely detected under ambient conditions. Surface-enhanced Raman spectroscopy (SERS) by 2D semiconductors relies on chemical (CM) enhancement driven by charge-transfer (CT) processes in bandgap alignment between molecules and substrates. Unfortunately, the low light absorption and weak conferment in the atomic-layer material limit the enhancement factor of Raman intensity (EFRI). Improving the utilization efficiency of excitation light is therefore essential for promoting SERS performance of 2D semiconductors.

A novel Corrax (CX) stainless steel was fabricated using the Selective Laser Melting (SLM) process. This study examined the influence of heat treatment processes on the microstructure and mechanical properties of CX stainless steel. Results indicate that SLM-fabricated CX samples mainly consist of martensite and residual austenite, with tensile strength and hardness of 1124 MPa and 337.

The pH stat in vitro lipolysis method is well established for evaluating lipid-based formulations (LBFs), however the absence of a simulated gastrointestinal transition may lead to an overestimation of drug precipitation particularly in the case of weakly basic drugs. This study aimed to compare the conventional pH-stat method with a pH-shift lipolysis approach by evaluating a diverse set of LBFs using nilotinib, a weakly basic model drug. Additionally, the study sought to assess in vitro-in vivo relationships (IVIVRs) and enhance understanding of the predictive capabilities of these models.

Unlike conventional phosphorus (P) recovery methods that rely on high P concentrations and/or chemical additives, this study presents a cost-effective approach for inducing the precipitation of phosphate salts by utilizing carefully selected feedstocks and optimal operating conditions. To test the feasibility of this approach, three 1.0-L upflow anaerobic sludge blanket (UASB) reactors were operated, fed with three types of synthetic feeds, consisting of glucose + sodium acetate (NaAc), glucose + bovine serum albumin (BSA), and glucose + urea as the organic matter sources.
