Tixagevimab/cilgavimab is a monoclonal antibody used to prevent coronavirus disease 2019 among immunocompromised hosts and maintained neutralizing activity against early omicron variants. Omicron BN.1 became a dominant circulating strain in Korea early 2023, but its susceptibility to tixagevimab/cilgavimab is unclear. We conducted plaque reduction neutralization test (PRNT) against BN.1 in a prospective cohort (14 patients and 30 specimens). BN.1 PRNT was conducted for one- and three-months after tixagevimab/cilgavimab administration and the average PRNT ND of each point was lower than the positive cut-off value of 20 (12.9 ± 4.5 and 13.2 ± 4.2, respectively, = 0.825). In the paired analyses, tixagevimab/cilgavimab-administered sera could not actively neutralize BN.1 (PRNT ND 11.5 ± 2.9, = 0.001), compared with the reserved activity against BA.5 (ND 310.5 ± 180.4). Unlike virus-like particle assay, tixagevimab/cilgavimab was not active against BN.1 in neutralizing assay, and would not be effective in the present predominance of BA.2.75 sublineages.

Chimeric antigen receptor T (CAR-T)-cell therapy is a promising resolution for solid tumors, but its corresponding clinical translation has been hindered by unsatisfactory therapeutic potency and severe cytokine release syndrome. Herein, tetracycline (Tet)-On inducible human epidermal growth factor receptor 1 (HER1)-targeted CAR-T (Tet-HER1-CAR-T) cells were engineered to enable spatially selective activation at tumor sites by doxycycline (Doxy), which is delivered by pH-responsive stealth liposomal calcium carbonate nanoparticles (Doxy@CaCO-PEG). Compared with the intravenous administration of conventional HER1-CAR-T cells and Tet-HER1-CAR-T cells activated by free Doxy, concurrent intravenous administration of Tet-HER1-CAR-T cells and Doxy@CaCO-PEG leads to the localized tumor activation of Tet-HER1-CAR-T cells and reduced systemic secretion of inflammatory cytokines.

Feline infectious peritonitis virus (FIPV) can cause an immune-mediated disease that is fatal to felines, but there is a lack of clinically effective protection conferred by vaccines. The methyltransferase (MTase) activity of the coronavirus nonstructural proteins nsp14 and nsp16 affects virulence, but there are no studies on the effect of nsp14 and nsp16 mutations affecting enzyme activity on the virulence of FIPV. In this study, we successfully rescued two mutant strains based on the previous infectious clone QS-79, named FIPV QS-79 dnsp14 and dnsp16, by mutating the MTase active sites of nsp14 (N415) and nsp16 (D129).
