Despite significant progress in the development of treatment options, melanoma remains a leading cause of death due to skin cancer. Advances in our understanding of the genetic, transcriptomic, and morphologic spectrum of benign and malignant melanocytic neoplasia have enabled the field to propose biomarkers with potential diagnostic, prognostic, and predictive value. While these proposed biomarkers have the potential to improve clinical decision making at multiple critical intervention points, most remain unvalidated. Clinical validation of even the most commonly assessed biomarkers will require substantial resources, including limited clinical specimens. It is therefore important to consider the properties that constitute a relevant and clinically-useful biomarker-based test prior to engaging in large validation studies. In this review article we adapt an established framework for determining minimally-useful biomarker test characteristics, and apply this framework to a discussion of currently used and proposed biomarkers designed to aid melanoma detection, staging, prognosis, and choice of treatment.

Acute promyelocytic leukemia (APL) is a medical emergency that needs immediate diagnosis and treatment. Podoplanin, a transmembrane glycoprotein that binds CLEC-2 on platelets, was recently demonstrated to be abnormally expressed in leukemic blasts in APL, as opposed to other forms of AML, in a study using thawed primary cells. This study aimed to explore and validate the diagnostic accuracy of measuring podoplanin expression by flow cytometry in the differential diagnosis of APL and other forms of acute myeloid leukemia (AML) as part of the diagnostic work-up of all cases suspected of AML in an academic hematology center.

Piezo-type mechanosensitive ion channel component 1 (Piezo1) is an evolutionarily conserved and multifunctional mechanosensitive ion channel protein that has emerged as a significant contributor to the pathogenesis of inflammatory bowel disease (IBD). Piezo1 plays a crucial role in regulating intestinal barrier integrity, immune responses, and the intestinal nervous system, thereby influencing disease progression. Its expression patterns correlate with disease severity and inflammatory markers in IBD patients, indicating its potential as a diagnostic and prognostic biomarker.

Background: This study investigates the impact of trimethylamine oxide (TMAO) on recurrent cerebral infarction in minor ischemic stroke (MIS).
Methods: A rat model was used, with dietary choline levels adjusted to vary TMAO levels. TMAO was quantified via liquid chromatography-mass spectrometry (LC-MS), and histological changes in brain and aortic tissues were analyzed using HE staining.

Non-hydrogenative para-hydrogen-induced polarization (nhPHIP) has proven a powerful tool for the enhanced NMR detection of several classes of metabolites in complex mixtures. Particularly, compounds carrying an α-amino acid motif have been previously detected and quantified in biological samples and natural extracts at submicromolar concentrations using 2D nhPHIP NMR spectroscopy. This technique is here applied for the first time in a semi-targeted metabolomics NMR study on urine from patients suffering from Pyridoxine-Dependent Epilepsy (PDE), currently diagnosed by the presence of dilute unique biomarkers.
