Poor drug penetration due to local skin thickening and microenvironmental redox imbalance remain a critical challenge in the treatment of epidermal diseases. Considering that oxidative stress due to the overproduction of Reactive Oxygen Species (ROS) in the skin microenvironment is one of the main mechanisms in the pathogenesis of psoriasis, we developed a ROS-sensitive transdermal drug delivery system to enhance topical treatment of psoriasis. Cross-linked cyclodextrin metal-organic framework (COF) containing curcumin (CUR) was dispersed in two types of dissolving polysaccharide microneedles (CPPAP-MNs-CUR@COF, CPPNP-MNs-CUR@COF) made from Codonopsis pilosula neutral polysaccharides (CPPNP) and acidic polysaccharides (CPPAP) for topical administration.

Acute ischemic stroke (AIS) is characterized by the abrupt onset of neurological dysfunction stemming from focal brain ischemia, confirmed through imaging evidence of infarction. In contrast, transient ischemic attack (TIA) manifests with neurological deficits in the absence of infarction, with imaging serving as the definitive diagnostic criterion. This study aims to assess the diagnostic value of combining non-high-density lipoprotein cholesterol (non-HDL-C) and blood pressure (BP) in differentiating AIS from TIA.

In cancer therapy, tumor cells can diminish their signals through mechanisms such as immune escape, thereby evading recognition and elimination by the immune system. Providing tumor signals to enhance the recognition of tumor sites is considered a crucial approach in cancer treatment. Inspired by the decoy-induced directed feeding of fish, we propose a biomimetic nanoparticle system for tumor signal amplification.

Background Posttranslational modifications of histone lysine (K) have integral connections with cell metabolism, and participate in the carcinogenesis of various cancers. This study focuses on evaluating the expression of histone H4 lys 5 lactylation (H4K5lac) and its clinical role in breast cancer (BC). Methods During this research, immunohistochemistry (IHC) and immunoblotting, utilizing a specific primary anti-L-lactyl-histone H4 (Lys 5) rabbit monoclonal antibody, were employed to assess H4K5lac expression in BC tissue chips.

Atrial fibrillation (AF) is the most prevalent arrhythmia in clinical practice, and obesity serves as a significant risk factor for its development. The underlying mechanisms of obesity-related AF remain intricate and have yet to be fully elucidated. We have identified FPR2 as a potential hub gene involved in obesity-related AF through comprehensive analysis of four transcriptome datasets from AF patients and one transcriptome dataset from obese individuals, and its expression is up-regulated in both AF and obese individuals.

Article Synopsis
  • Inflammatory bowel disease (IBD) is marked by chronic inflammation in the digestive tract and involves blood vessel formation (angiogenesis) as a key factor in its progression.
  • Ginsenoside Rg3 from ginseng has anti-angiogenic effects but suffers from poor absorption in the body, prompting the development of a new delivery system using a thermosensitive hydrogel called Gel-Rg3, made with Pluronic F-127 and F-68.
  • Gel-Rg3 demonstrated better safety and effectiveness against IBD in lab tests and mouse models by promoting drug retention in the intestine and inhibiting harmful blood vessel formation, offering new treatment strategies for managing IBD.

Myricetin, a natural flavonoid found in various foods, was investigated for its antiviral effect against transmissible gastroenteritis virus (TGEV). This α-coronavirus causes significant economic losses in the global swine industry. The study focused on the papain-like protease (PLpro), which plays a crucial role in coronavirus immune evasion by mediating deubiquitination.

Pullorum disease (PD) is a bacterial infection caused by () that affects poultry. It is highly infectious and often fatal. Antibiotics are currently the mainstay of prophylactic and therapeutic treatments for PD, but their use can lead to the development of resistance in pathogenic bacteria and disruption of the host's intestinal flora.

Article Synopsis
  • - Chuanminshen violaceum (CV) is used in traditional medicine to improve lung function, aid digestion, reduce phlegm, and detoxify, with its active polysaccharides not thoroughly understood from its stems and leaves.
  • - This study aimed to optimize the extraction of polysaccharides from CV using ultrasound, analyze their structure, and evaluate their anti-inflammatory and antioxidant properties with a focus on their mechanisms of action.
  • - Researchers successfully isolated a polysaccharide (CVP-AP-I) that was primarily pectic, demonstrated a molecular weight of 35.34 kDa, and showed potential in reducing inflammation and oxidative stress in intestinal cells following exposure to LPS.

Paeoniae Radix alba is used in Traditional Chinese Medicine for the treatment of gastrointestinal disorders, immunomodulatory, cancer, and other diseases. In the current study, the yield of Paeoniae Radix alba polysaccharide (PRP) was significantly increased with optimal ultrasound-assisted extraction compared to hot water extraction. Further, an acidic polysaccharide (PRP-AP) was isolated from PRP after chromatographic separation and was characterized as a typical pectic polysaccharide with side chains of arabinogalactans types I and II.

Lead and cadmium are foodborne contaminants that threaten human and animal health. It is well known that lead and cadmium produce hepatotoxicity; however, defense mechanisms against the co-toxic effects of lead and cadmium remain unknown. We investigated the mechanism of autophagy (defense mechanism) against the co-induced toxicity of lead and cadmium in rat hepatocytes (BRL-3A cells).

Drug delivery systems based on nanoparticles still face challenges of low efficacy and an inability to track treatment effects in tumor therapy due to biological barriers. This limitation hinders clinicians' ability to determine treatment effects and proper drug dosages, thus, ultimately impeding the further application and transformation of nanoplatforms. To address this challenge, an all-in-one nanoplatform for therapy and imaging is proposed.

Mitochondria in breast cancer play a critical role in survival and adaptation to dynamic environments. Thus, targeting mitochondria emerges as a promising therapeutic strategy for breast cancer. However, the adaptive unfolded protein response in mitochondria (UPR) due to mitochondrial unspecific distribution might contribute to diminished therapeutic outcomes.
