Inadequate antigen presentation by MHC-I in tumor microenvironment (TME) is a common immune escape mechanism. Here, we show that glycine decarboxylase (GLDC), a key enzyme in glycine metabolism, functions as an inhibitor of MHC-I expression in EGFR-activated tumor cells to induce immune escape by a mechanism independent of its enzymatic activity. Upon EGFR activation, GLDC is phosphorylated by SRC and subsequently translocated to the nucleus in human NSCLC cells.

Although dynamical systems models are a powerful tool for analysing microbial ecosystems, challenges in learning these models from complex microbiome datasets and interpreting their outputs limit use. We introduce the Microbial Dynamical Systems Inference Engine 2 (MDSINE2), a Bayesian method that learns compact and interpretable ecosystems-scale dynamical systems models from microbiome timeseries data. Microbial dynamics are modelled as stochastic processes driven by interaction modules, or groups of microbes with similar interaction structure and responses to perturbations, and additionally, noise characteristics of data are modelled.

Sodium-glucose Cotransporter 2 (SGLT-2) inhibitors are oral antidiabetic drugs that were developed for the treatment of patients with diabetes mellitus and are now also approved for treating chronic heart failure and chronic kidney disease. Primary microcephaly, a rare congenital condition characterized by reduced brain size, occurs due to impaired neurogenesis during brain development. Through whole-exome sequencing, we identified compound heterozygous loss-of-function mutations in CENTRIN 3 (CETN3) in a 5-year-old patient with primary microcephaly. As CETN3 has not been previously linked to microcephaly, we investigated its potential function in neurodevelopment in human pluripotent stem cell-derived cerebral organoids.

Selected were a large subunit catalytic (Met-116-Leu) mutation that increases the k of varying plant Rubiscos by 25% to 40% and a solubility (Ala-242-Val) mutation that improves plant Rubisco biogenesis in E. coli 2- to 10-fold.

Purpose: To evaluate the preclinical efficacy and safety of transarterial chemoembolization (TACE) using doxorubicin-loaded biocompatible cellulose nanoparticles in a rabbit VX2 liver tumor model.
Materials And Methods: Following institutional animal care committee approval, 23 rabbits with VX2 liver tumors were randomized into three groups: Group A (n = 9) received doxorubicin-loaded cellulose nanoparticles with ethiodized oil; Group B (n = 9) received doxorubicin with ethiodized oil; and Group C (n = 5) served as untreated controls. Tumor size was monitored via ultrasound for 4 weeks, and serum liver enzymes (aspartate transaminase and alanine transaminase) were measured on days 1, 3, and 7 to assess hepatotoxicity.

Research conducted over the last 15 years indicates that cAMP is generated not just from the plasma membrane but also from intracellular compartments, particularly in endosomes, where receptors are redistributed during the endocytosis process. This review centers on the parathyroid hormone type 1 receptor (PTHR) as a model for a peptide hormone GPCRs that generates cAMP from various locations with distinct duration and pharmacological effectiveness. We discuss how structural dynamics simulations aid in designing ligands that induce cAMP location bias, ultimately answering how the spatiotemporal generation of cAMP affects pharmacological responses mediated by the PTHR.

A ketogenic diet (KD) has shown promise as an adjunctive therapy for neurological and neuropsychiatric disorders, including bipolar disorder and major depressive disorder (MDD). We examined tolerance for a KD in young adults with MDD and assessed symptoms of depression and metabolic health. Students (n = 24) with a confirmed diagnosis of MDD at baseline receiving standard of care counseling and/or medication treatment were enrolled in a 10-12 week KD intervention that included partial provision of ketogenic-appropriate food items, frequent dietary counseling, and daily morning tracking of capillary R-beta-hydroxybutyrate (R-BHB).

Ether-based electrolytes are widely acknowledged for their potential to form stable solid electrolyte interfaces (SEIs) for stable anode performance. However, conventional ether-based electrolytes have shown a tendency for cation-solvent co-intercalation phenomena on graphite electrodes, resulting in lower capacity and higher voltage platforms compared to those of neat cation insertion in ester-based electrolytes. In response, we propose the development of weakly solvating ether solvents to weaken the interaction between cations and solvents, thereby suppressing co-intercalation behavior.
