The accurate characterization of glycosyl linkages in complex carbohydrates is always a challenging task due to the occurrence of isomer diversity and the unavailability of known standards. Because of the significant difference between head and tail in structures, this article built a complete methylation analysis library based on partially methylated aldononitrile acetates (PMANs) for facilitated identification of complex glycosidic linkages in polysaccharides. This library covered full EI-MS spectra and detailed fragmentation behaviors of corresponding PMANs ascribed to 46 aldohexoses, 32 aldopentoses and 16 deoxyhexoses. Furthermore, the elute behaviors of these PMANs are summarized on two capillary columns ascribed to complementary polarities. It is quite benefit to elucidate the glycosyl linkages of polysaccharides for inexperienced researchers in this field. The newly established PMAN library has been conformed to be an effective and alternative strategy for glycosyl linkage analysis of polysaccharides.

The current study aims to examine the efficacy of a novel esterase derived from an endophyte fungus in decolorizing diverse dyes, focusing on its production, purification, optimization, and characterization.
Results: Trichoderma afroharzianum AUMC16433, a novel fungal endophyte with esterase-producing ability, was first detected from the cladodes of Opuntia ficus indica by ITS-rRNA sequencing.

The PR10 (Pathogenesis-Related Protein 10) family plays a crucial role in plant defense and growth regulation, with unique hydrophobic cavities that bind various ligands, including phytohormones and alkaloids. The tumor microenvironment (TME) plays a pivotal role in cancer progression, though the molecular regulators governing its immunosuppressive properties remain incompletely characterized. In this study, we identify Makorin-2 (MKRN2) as a novel modulator of TME remodeling through integrated analyses of genetically engineered mouse models and human clinical data. Utilizing MKRN2 knockout mice, we observed significantly accelerated tumor growth compared to wild-type control, which was associated with profound alterations in immune cell composition, especially M2 macrophages.

Objective: Major depressive disorder (MDD) is among the most prevalent and debilitating mental health conditions worldwide. This study aims to investigate the bidirectional causal relationship between immune cells and MDD using Mendelian randomization (MR) analysis and determine whether metabolites mediate this relationship.
