The effector functions of CD8 T cell significantly influence the immunosuppressive microenvironment in hepatocellular carcinoma (HCC), which is intricately associated with HCC prognosis. Nevertheless, a comprehensive investigation into the relationship between CD8 T cell immune-related genes and HCC prognosis remains lacking. This study aimed to construct a prognostic model for HCC using CD8 T cell immune-related genes to provide insights for clinical management and prognosis. A prognostic model was constructed by incorporating 16 CD8 T cell-specific immune-related genes, yielding area under the curve (AUC) values of 0.821, 0.796, and 0.784 for the prediction of 1-year, 3-year, and 5-year survival, respectively, receiver operating characteristic (ROC) curve analysis. The qRT-PCR results showed that the mRNA levels of PTMA, RAC1, HSPD1, HSP90AA1, and TANK were significantly higher in HCC cells compared to normal cells ( < 0.05). Further analysis focused on the TANK gene, which was significantly upregulated in HCC tissues ( < 0.05). The CCK-8 and wound healing assays revealed a significant decrease in both the cell proliferation rate and wound-healing rate in the TANK-deficient group compared with the control group ( < 0.05). These findings may offer new insights into the clinical treatment and prognostic evaluation of HCC.

Lymphotoxin β receptor (LTβR/TNFRSF3) signaling plays a crucial role in immune defense. Notably, LTβR-deficient (LTβR) mice exhibit severe defects in innate and adaptive immunity against various pathogens and succumb to infection. Here, we investigated the bone marrow (BM) and peritoneal cavity (PerC) compartments of LTβR mice during infection, demonstrating perturbed B-cell and T-cell subpopulations in the absence of LTβR signaling.

Contagious bovine pleuropneumonia (CBPP), caused by subsp. (Mmm), is a devastating cattle disease with high morbidity and mortality, threatening cattle productivity in Sub-Saharan Africa and potentially in parts of Asia. Cross-border livestock trade increases the risk of CBPP introduction or reintroduction.

Chimeric antigen receptor T-cell (CAR-T) therapies have demonstrated clinical efficacy in treating haematological malignancies, resulting in multiple regulatory approvals. However, there is a need for robust manufacturing platforms and the use of GMP-aligned reagents to meet the clinical and commercial demands. This study investigates the impact of serum/xeno-free medium (SXFM) and cytokine supplementation on CAR-T cell production in static and agitated culture systems, using 24-well plate G-Rex vessels and 500 mL stirred tank bioreactors (STRs), respectively.

Immune checkpoint inhibitors (ICIs) can re-active the immune response and induce a complete response in mismatch repair-deficient and microsatellite instability-high (dMMR/MSI-H) colorectal cancer (CRC). However, most CRCs exhibit proficient mismatch repair and microsatellite stable (pMMR/MSS) phenotypes with limited immunotherapy response because of sparse intratumoral CD8 T-lymphocyte infiltration. Cellular senescence has been reported to involve immune cell infiltration through a senescence-associated secretory phenotype (SASP).
