Article Abstract

Dearomatization of planar aromatics offers unparalleled opportunities for the construction of three-dimensional stereochemical frameworks. However, the inherent instability of dearomatized intermediates, particularly bearing labile C(sp)-heteroatom bonds, has hindered progress due to competing rearomatization pathways. By capitalizing on arene dearomatization and transforming the rearomatization predicament into a new opportunity, we herein present a stability-driven catalytic asymmetric dearomatization/rearomatization (CADA/RA) cooperative strategy to resolve racemic -stereogenic compounds. Through the systematic engineering of phenolic substrates, bench-stable -site dearomatized products with dual C(sp)-P and C(sp)-Cl bonds were isolated. Chiral Lewis acid catalysis enabled kinetic resolution (KR) and parallel kinetic resolution (PKR), achieving full enantiomeric separation with exceptional stereocontrol across diverse substrates. Mechanistic analysis revealed competing substrate- and catalyst-controlled pathways for stereochemical precision. Postsynthetic chemoselective - or -liberation furnished enantioenriched -chiral compounds. This method transforms the challenges of dearomatization and rearomatization into opportunities for stereochemical complexity generation, offering a robust platform to synthesize -chiral building blocks and ligand precursors.

Purpose: We report the efficacy and safety of voretigene neparvovec (VN) as an adeno-associated viral vector-based gene therapy for Japanese patients with inherited retinal dystrophy caused by biallelic pathogenic variants (-retinopathy).

Design: Open-label, single arm, phase III clinical trial.

Participants: Four subjects were recruited based on the following criteria: (1) a clinical and molecular genetic diagnosis of -retinopathy; (2) age ≥4 years; (3) a best-corrected VA (BCVA) worse than 20/60 or a visual field (VF) <20° by a III4e isopter or equivalent; and (4) sufficient viable retinal cells by OCT or ophthalmoscopy.

Liposomal carriers, used for site-specific drug delivery, are being investigated for diagnostic approaches by replacing the therapeutic with an imaging contrast agent, exploring potential for selective treatment planning. There remains a critical need to improve assessment of biodistribution, stability, and clearance kinetics of liposomal carriers. This pilot study presents a multimodal approach in which liposome-encapsulated J-aggregated indocyanine green (ICG) dye (Lipo-JICG) is imaged with high spatial resolution using both photoacoustic (PA) imaging, to assess the absorbance characteristics of JICG and monomeric ICG, and cryofluorescence tomography (CFT), to measure ICG fluorescence.

Spectro-Electrochemical Insights into Electrocatalytic CO Reduction in Acidic Media through Model Catalyst Design.

Electrocatalytic CO reduction (eCOR) under acidic conditions is the game changer of resourceful CO utilization owing to the alleviated carbon loss but faces severe competition from the hydrogen evolution reaction (HER) that greatly curtails the electric current efficiency. Leveraging the eCOR side of the teeterboard calls for a fundamental understanding of the triphasic electrode process involving a complex arrangement of electric double layers (EDLs). Herein, a series of model catalysts with tailored cavernous parameters are fabricated to geometrically and spectroscopically decipher the competing HER and eCOR processes that engage different proton sources.

The precise spatial and temporal regulation of gene expression through enhancer-promoter (E-P) interactions represents a fundamental mechanism underlying cellular differentiation and organismal development in multicellular eukaryotes. Despite extensive studies on enhancer-mediated gene regulation, a systematic understanding of how specific E-P configurations affect transcriptional dynamics remains incomplete. Recent advances in live-imaging, single-cell assays, and chromatin conformation capture technologies have enabled unprecedented insights into these dynamic regulatory processes by providing temporal resolution and single-cell specificity that complement traditional population-based approaches.
