Article Abstract

Emamectin benzoate (EB), a derivative of avermectin, is the primary insecticide used to control the fall armyworm (FAW) in China. However, the specific molecular targets of EB against FAW remain unclear. In this study, we cloned the glutamate-gated chloride channel (GluCl) gene, which is known to be a primary molecular target for avermectin. We first investigated the transcript levels of SfGluCl in FAW and found that the expression level of SfGluCl in the head and nerve cord was significantly higher than that in other tissues. Furthermore, we found that the expression level of SfGluCl was significantly higher in eggs than that in other developmental stages, including larvae, pupae, and adults. Additionally, we identified three variable splice forms of SfGluCl in exons 3 and 9 and found that their splice frequencies remained unaffected by treatment with the LC of EB. RNAi mediated knockdown of SfGluCl showed a significant reduction of 42% and 65% after 48 and 72 h of dsRNA feeding, respectively. Importantly, knockdown of SfGluCl sifgnificantly reduced LC and LC EB treatment induced mortality of FAW larvae by 15% and 44%, respectively, compared to the control group feeding by dsEGFP. In contrast, there were no significant changes in the mortality of FAW larvae treated with the control insecticides chlorantraniliprole and spinetoram. Finally, molecular docking simulations revealed that EB bound to the large amino-terminal extracellular domain of SfGluCl by forming five hydrogen bonds, two alkyl hydrophobic interactions and one salt bridge. These findings strongly suggest that GluCl may serve as one of the molecular targets of EB in FAW, shedding light on the mode of action of this important insecticide.

There is a need to improve the discovery of new drugs for neglected tropical diseases (NTDs), as the lack of financial incentives has slowed their development. Currently, ivermectin and moxidectin are used in the management of onchocerciasis. We present a proof-of-concept study based on computational methods to find anti-infectives that can be repurposed or serve as lead compounds for onchocerciasis.

Root-knot nematodes (Meloidogyne incognita) present a significant threat to global agriculture, and the development of multi-drug resistance in these nematodes exacerbates this problem. Benzothiazole, a heterocyclic compound has been reported as a potential nematicide, however, its mode of action is not fully understood. This study aims to elucidate the nematicidal mechanism of benzothiazole against M.

The rice stem borer, Chilo suppressalis (Walker), is a major pest of rice crops. It is commonly managed by pesticides, several of which act on cys-loop ligand-gated ion channels (cysLGICs). In insects, the cysLGIC superfamily consists of receptors that mediate fast excitatory or inhibitory synaptic transmission in the nervous system as well as performing non-neuronal roles.

Background: Current regulatory requirements for European marketing authorization of canine and feline ectoparasiticides include dose confirmation studies conducted with European isolates of each ectoparasite species indicated in the proposed labeling, in addition to any studies conducted against non-European ectoparasite isolates. This regulatory requirement may be deemed unnecessary if no significant differences exist among laboratory ectoparasite isolates obtained from various countries. Between-country interchangeability was examined through various comparative studies by using Ctenocephalides felis fleas and Rhipicephalus sanguineus sensu lato ticks sourced from multiple countries.

Although the present use of pesticides in plant protection has limited the occurrence and development of plant diseases and pests, resistance to pesticides and their environmental and health hazards indicates an urgent need for new active ingredients in plant protection products. Recently synthesized coumarin-1,2,4-triazole hybrid compounds have been proven effective against plant pathogenic fungi and safe for soil-beneficial bacteria. , the common fruit fly, has been used as a model organism for scientific research.
