Synthesis and Vasorelaxant Activity of Nitrate-Coumarin Derivatives.

Article Abstract

Due to the need for new chemical entities for cardiovascular diseases, we have synthesized a new series of nitrate-coumarins and evaluated their vasorelaxant activity in contraction-relaxation studies using rat aorta rings precontracted with phenylephrine or by depolarization with a high concentration of potassium chloride. Four of the new compounds were able to relax smooth vascular muscle with a similar profile and potency to glyceryl trinitrate (IC =12.73 nM) and sodium nitroprusside (IC =4.32 nM). Coumarin-7-yl-methyl nitrate (4), the best compound within the series, was able to relax smooth vascular muscle in the low nanomolar range (IC =1.92 nM). The mechanisms of action have been explored, being the activation of sGC and the opening of K channels involved. Our studies indicate that the new nitrate derivatives are reversible and not deleterious for aortic rings, suggesting that these compounds have a potential interest for the development of new and highly efficient vasodilator drugs.

This study examined whether consistent sleep could improve vascular function and mitigate the negative effect of 25-hour total sleep deprivation. Enoxaparin sodium (ES), a low molecular weight heparin derivative, has recently been recognized for its diverse biological activities. In particular, the ability of heparin to modulate inflammation has been utilized to enhance the biocompatibility of bone implant materials. In this study, we utilized poly (methyl methacrylate) (PMMA), a drug loading bone implant material, as a matrix and combined this with enoxaparin sodium (ES) to create enoxaparin sodium PMMA cement (ES-PMMA) to investigate the regulatory effects of ES on inflammatory responses in bone tissue from an animal model.

Sequential treatment for diabetic foot ulcers in aortic dissection patients: a case report.

Diabetic foot ulcers (DFUs) represent a prevalent complication of diabetes, with a lifetime risk ranging from 15% to 25% among diabetic patients. Research indicates that anticoagulation plays a crucial role in the management of newly diagnosed cases of diabetic lower extremity atherosclerotic obliterative disease. However, in the present case, the patient developed dry gangrene in both toes after receiving vasodilator drugs during an emergency intervention for sudden aortic dissection.
