Long QT syndromes (LQTS) are heritable diseases characterized by prolongation of the QT interval on an electrocardiogram, which often leads to syncope and sudden cardiac death. Here we report the generation of induced pluripotent stems (iPS) cells from two patients with LQTS type 3 carrying a different point mutation in a sodium channel Nav1.5 (p.V240M and p.R535Q) and functional characterization of cardiomyocytes (CM) derived from them. The iPS cells exhibited all characteristic properties of pluripotent stem cells, maintained the disease-specific mutation and readily differentiated to CM. The duration of action potentials at 50% and 90% repolarization was longer in LQTS-3 CM as compared to control CM but this difference did not reach statistical significance due to high variations among cells. Sodium current recordings demonstrated longer time to peak and longer time to 90% of inactivation of the Na(+) channel in the LQTS-3 CM. This hints at a defective Na(+) channel caused by deficiency in open-state inactivation of the Na(+) channel that is characteristic of LQTS-3. These analyses suggest that the effect of channel mutation in the diseased CM is demonstrated in vitro and that the iPS cell-derived CM can serve as a model system for studying the pathophysiology of LQTS-3, toxicity testing and design of novel therapeutics. However, further improvements in the model are still required to reduce cell-to-cell and cell line-to-cell line variability.

Introduction: The α-adrenoceptor (αAR) is involved in the physiopathology of the central nervous system (CNS), but its function in the adult male rat locus coeruleus (LC) has not been fully studied. We aimed to characterize the role of the αAR in the regulation of the firing rate (FR) of LC neurons and to describe the signaling pathways involved.
Methods: We measured, through single-unit extracellular recordings of LC neurons from adult male rats were used to measure the effect of adrenergic agonists in the presence and absence of adrenergic antagonists or inhibitors of several signalling pathways.

Tetrodotoxin (TTX), the pufferfish toxin, has the potential to cause fatal food poisoning because of its potent voltage-gated sodium channel (Na) blocking activity. 4-epiTTX, 11-norTTX-6(S)-ol, and 11-oxoTTX are the major TTX analogues found in marine animals; thus, their chemical properties and biological activities should be determined. In this study, these three TTX analogues were purified to a high level (purity >97%) from pufferfish and newts.

Achieving stable and continuous monitoring of signals of numerous single neurons in the brain faces the conflicting challenge of increasing the microelectrode count while minimizing cross-sectional shank dimensions to reduce tissue damage, foreign-body-reaction and maintain signal quality. Passive probes need to route each microelectrode individually to external electronics, thus increasing shank size and tissue-damage as the number of electrodes grows. Active complementary metal-oxide-semiconductor (CMOS) probes overcome the limitation in electrode count and density with on-probe frontend, addressing and multiplexing circuits, but current probes have relatively large shank widths of 70 - 100 μm.

Here, intramolecular hydrogen bond (IMHBs)-induced rigidity is used for the first time to synthesize macrocyclic arenes. Calix[]azanediyldibenzoates (C[]A, where = 3, 4, or 5) are synthesized through a one-step condensation reaction between dimethyl 2,2'-azanediyldibenzoate and paraformaldehyde. Compared to the monomer, the macrocycles exhibit a fast and significant acidochromic response due to the intramolecular charge transfer that is boosted by the synergistic effect of their adsorption and protonation.
