According to the Food and Agriculture Organization of the United Nations (FAO), pests reduce global crop production by 14% annually. The growing challenge of pest resistance, coupled with the relatively low success rates of pesticides, has prompted researchers to shift their attention towards the accurate evaluation of insecticide lead. In contrast to in vitro methods of structural similarity or target affinity, the 'insecticide-likeness' approach emphasises the in vivo biological effects of compounds, thereby constructing precise and comprehensive evaluation rules.

Vascular dementia (VaD) is a neurodegenerative disorder characterized by chronic oxygen insufficiency, leading to the generation of oxygen-free radicals, inflammatory responses, disturbances in iron metabolism, lipid peroxidation, and other pathological changes that disrupt intracellular homeostasis. These processes ultimately lead to neuronal death and cognitive dysfunction. Normal neurological functions depend on the capacity of the iron homeostatic system to regulate the balance of oxidative states.

The chiral carbocyclic and heterocyclic spirane scaffolds are widely present in natural products and pharmaceutical molecules. Efficient, concise, and enantioenriched synthesis of these structures is a significant objective in both academic and industrial fields. Here, we design and synthesize a series of chiral SPA-PNN ligands, which are used to kinetically resolve racemic spirocyclic 1,3-diketones and their analogs via Ir-catalyzed asymmetric direct hydrogenation to afford the corresponding chiral spirocyclic 1,3-diketones and chiral hydroxyspiroketones.

Enamides are distinctive amphiphilic synthons that can be strategically incorporated into cyclization reactions. The iminium species generated from enamides via nucleophilic addition or substitution are capable of engaging in further electrophilic additions or isomerization processes. Exploiting the multiple reactivities of enamides facilitates the development of diverse cyclization modes that provide entries to various -heterocycles, some of which serve as key structural motifs in natural alkaloids.

Neuroplastin 65 (NP65) is a synapse-enriched glycoprotein in the central nervous system and is implicated in synaptic plasticity. In the present study, we found that NP65 knockout (NP65 KO) mice exhibit impaired visual function, including reductions in the amplitude of b-wave in scotopic flash electroretinogram (fERG), the amplitude of N1 and P1 waves in flash visual evoked potentials (fVEP), and the constriction rate in pupillary light reflexes (PLR). In wild-type (WT) mice, NP65 is specifically enriched in the synaptic ribbon (SR) of ribbon synapses labeled by Ribeye in the retina.

The all-carbon quaternary stereogenic center of oxindoles is a crucial structural element of a broad spectrum of indole alkaloids, imparting these molecules with rigid three-dimensional configurations essential for their biological activities. Here, we present a catalytic asymmetric α-ethynylation reaction of oxindoles taking advantage of the catalysis of a spiropyrrolidine amide (SPA) triazolium. This transformation enables the enantioselective construction of the C3 quaternary carbon stereocenter of oxindoles while introducing a versatile ethynyl functionality.

Chiral phospholane ligands and catalysts have been widely applied in asymmetric catalysis and synthesis. However, the construction of the chiral phospholane skeleton remains challenging and primarily relies on the use of chiral auxiliaries or resolution. In this work, a highly enantioselective and diastereoselective synthesis of P-stereogenic phospholane oxides has been achieved through a cobalt-catalyzed desymmetric hydroalkylation strategy.

Cerebral ischemia-reperfusion injury (CIRI) is a complex pathophysiological process faced by brain tissues after ischemic stroke treatment, which involves mechanisms of inflammatory response, oxidative stress and apoptosis, and severely affects treatment outcome. Lipocalin-2 (LCN2), an acute-phase protein, is significantly up-regulated after CIRI and promotes neural repair by enhancing astrocyte phagocytosis, but its over-activation may also trigger secondary inflammation and demyelination injury. LCN2 also plays a key role in neuroinflammation regulation by regulating the polarization state of astrocytes and the release of inflammatory factors, and may affect the integrity of the blood-brain barrier and a variety of pathologic injury processes.

The Lewis acid-catalyzed tittle reaction of 1,3-dicycloalkenlidine ketones is recognized as so far the shortest and most effective 1-step method for construction of angular tricyclic scaffolds, which are extensively found in bioactive terpenoids. Here, a further kinetic study of this reaction with 30 reaction examples is carried out using in situ IR technology and DFT calculation. That enables the creation of well-fitted linear relationships of lnk/(ΔG/T), ΔG/ΔG, ln(k/k)/σ, reflecting the structure's effect on reactivity/selectivity, and validating the reaction mechanism.
