Introduction: Providing precise oncologic treatment for patients with refractory solid tumor is still an unmet need in clinical practice. This study aimed to assess whether treatments recommended by a molecular tumor board (MTB) can improve clinical outcomes in patients with refractory solid tumors.

Methods: We screened all patients with refractory solid tumor during the period from 2017 to 2022 at the authors' center.

Evidence-guided regimens for advanced gastric cancer (AGC) in patients with performance status 2 (PS 2) are limited. Here, we proposed a structured therapeutic framework termed "performance status-matched strategy", and further conducted the APICAL-GC trial (NCT04278222). This open-label, single-arm phase II study evaluated the efficacy and safety of anlotinib combined with toripalimab among 24 treatment-naïve AGC patients with PS 2.

In the era of precision medicine, it has been increasingly recognized that individuals with a certain disease are complex and different from each other. Due to the underestimation of the significant heterogeneity across participants in traditional "one-size-fits-all" trials, patient-centered trials that could provide optimal therapy customization to individuals with specific biomarkers were developed including the basket, umbrella, and platform trial designs under the master protocol framework. In recent years, the successive FDA approval of indications based on biomarker-guided master protocol designs has demonstrated that these new clinical trials are ushering in tremendous opportunities.

The combination of immunotherapy and antiangiogenic agents for the treatment of refractory solid tumor has not been well investigated. Thus, our study aimed to evaluate the efficacy and safety of a new regimen of anlotinib plus PD-1 inhibitor to treat refractory solid tumor. APICAL-RST is an investigator-initiated, open-label, single-arm, phase II trial in patients with heavily treated, refractory, metastatic solid tumor.

The epidermal growth factor receptor (EGFR) tyrosine kinase inhibitors (TKIs) dramatically improve the clinical outcomes of non-small cell lung cancer (NSCLC) patients harboring EGFR -sensitive mutations. Despite the remarkable efficacy of first-and second-generation EGFR TKIs, disease relapse is inevitable. EGFR T790M mutation is a primary contributor to the acquired resistance to first- and second-generation EGFR TKIs.

Article Synopsis
  • Tumor mutation burden (TMB) is linked to how effectively patients with advanced gastric cancer respond to immunotherapy and chemotherapy; this study investigates its prognostic value.
  • Among the 155 patients analyzed, about 18.7% were classified as TMB-high, and those patients exhibited poorer overall (OS) and progression-free survival (PFS) compared to TMB-low patients—indicating that higher TMB is associated with worse outcomes.
  • The study suggests that the unfavorable prognosis in TMB-high patients may be due to lower metabolic activity, as indicated by differentially expressed genes associated with TMB status.

Hepatocellular carcinoma (HCC) is defined as a universal malignancy while radiation therapy is the effective treatment for it. This study validated the mechanism of long non-coding RNA (lncRNA) colorectal neoplasia differentially expressed gene (CRNDE) in radiation resistance in HCC. LncRNA CRNDE upregulation was detected in HCC cells.

Breast cancer is the most common type of cancer amongst women worldwide, and numerous microRNAs (miRNAs/miRs) are involved in the initiation and progression of breast cancer. The aim of the present study was to identify hub miRNAs and determine the underlying mechanisms regulated by these miRNAs in breast cancer. Breast invasive carcinoma transcriptome data (including mRNAs and miRNAs), and clinical data were acquired from The Cancer Genome Atlas database.

The present study aimed to evaluate the effect of liver metastases on the efficacy from the combination of PD-1/PD-L1 inhibitor with chemotherapy as first-line treatment in lung cancer using the meta-analysis. A total of 8 randomized controlled trials (RCTs) were included. In patients without liver metastases, PD-1/PD-L1 inhibitor plus chemotherapy could decrease the risk of progression by 40% and risk of death by 29% (HR = 0.

Article Synopsis
  • Chinese medicine is complex, mostly derived from plants or animals, and features unique chromatographic fingerprints that reflect its dynamic components.
  • A new mathematical model based on total quantum statistical moment (TQSM) has been developed to study the pharmacokinetics of multiple components in Chinese medicine, using Buyang Huanwu Decoction as a test case.
  • Key metabolic parameters were determined, indicating that 95% of the compounds are metabolized and eliminated from the body within approximately 96 hours, validating the effectiveness of the TQSM model in understanding the medicine's dynamics.

Electrospinning has been widely used as a nanofiber fabrication technique. Its simple process, cost effectiveness and versatility have appealed to materials scientists globally. Pristine polymeric nanofibers or composite nanofibers with dissimilar morphologies and multidimensional assemblies ranging from one dimension (1D) to three dimensions (3D) can be obtained from electrospinning.

Electrospinning (e-spinning) devices and electrospun (e-spun) ultrathin fibers have shown promising applications in various fields. However, the poor portability of conventional e-spinning devices limits some potential applications especially in the case without a plug (electricity supply). Consequently, great efforts have been made to modify e-spinning setups with good portability.

A conventional melt electrospinning setup usually needs a large, heavy high-voltage power supply and cannot work without a plug (electricity supply). In this article, we report a new melt electrospinning setup based on a small hand-operated Wimshurst generator, which can avoid electrical interference between the high-voltage spinning system and the heating system, and make the setup very portable and safe. Poly(lactic acid) (PLA) and polycaprolactone (PCL) fibers with diameters of 15-45 μm were fabricated successfully by using this apparatus.
