Background: Lysine-specific demethylase 1 (LSD1) is a histone demethylase and regulator of differentiation, including in cancer. A neuronal-specific isoform of LSD1-LSD1+8a-has been shown to play a key role in promoting neuronal differentiation in the developing brain. We previously determined that LSD1+8a transcripts were detected in an aggressive subtype of prostate cancer harboring a neuronal program-neuroendocrine prostate cancer (NEPC)-but not in prostate adenocarcinomas harboring a glandular program.

  • Biallelic loss of CDK12 is linked to a specific subtype of metastatic castration-resistant prostate cancer (mCRPC), raising questions about its role in cancer development versus exposing drug targets.
  • Research shows that loss of CDK12 leads to early cancer-like changes and enhances cancer cell growth when combined with mutations in other genes like Trp53, while it inhibits tumor growth in the absence of another tumor suppressor gene, Pten.
  • CDK12 loss causes genomic instability and makes tumors sensitive to treatments targeting another protein, CDK13, highlighting CDK12 as a crucial tumor suppressor and suggesting new therapeutic approaches for CDK12-mutant mCRPC.

Article Synopsis
  • - A two-step method combining softened spark microarc oxidation technology was developed to create composite coatings with varying compositions on Ti alloys, focusing on the Ti6Al4V alloy. - The study revealed that softened spark discharges favored nucleation at the coating/substrate interface and had a lower process voltage than the anodic oxide film's breakdown voltage, impacting anion deposition in the electrolyte. - The results indicated that the softened sparks enhanced rutile phase formation and thickness growth of the coating, with key elements like Ca and P mostly found in the external porous layer, while the initial micro/nanoporous structures of the coatings remained largely unaffected.

  • Biallelic loss of the gene encoding cyclin-dependent kinase 12 (CDK12) is linked to a distinct type of advanced prostate cancer, known as metastatic castration-resistant prostate cancer (mCRPC), though its role in cancer development and treatment response is still being investigated.
  • Research using mouse models shows that loss of CDK12 can lead to early-stage cancer changes and increased immune response, as well as promote tumor growth in lab settings when paired with other genetic modifications.
  • CDK12 mutations make tumors more responsive to certain immunotherapies and therapies targeting related kinases, suggesting that CDK12 acts as a tumor suppressor and emphasizing the potential for new treatment strategies focusing on related gene interactions in mutant m

Birt-Hogg-Dubé (BHD) syndrome is associated with an increased risk of multifocal renal tumors, including hybrid oncocytic tumor (HOT) and chromophobe renal cell carcinoma (chRCC). HOT exhibits heterogenous histologic features overlapping with chRCC and benign renal oncocytoma, posing challenges in diagnosis of HOT and renal tumor entities resembling HOT. In this study, we performed integrative analysis of bulk and single-cell RNA sequencing data from renal tumors and normal kidney tissues, and nominated candidate biomarkers of HOT, L1CAM, and LINC01187 , which are also lineage-specific markers labeling the principal cell and intercalated cell lineages of the distal nephron, respectively.
