Background: High-resolution respirometry is commonly used in skeletal muscle research and exercise science to measure mitochondrial respiratory function in both permeabilized muscle fibers and isolated mitochondria. Due to the low throughput and high cost of the most used respirometer, the Oroboros 2k (O2k), multiple experiments are often conducted within the same chamber in short succession. Despite this, no methodological consideration has been given for the potential contamination of inhibitors, used to investigate the contribution of specific complexes within the electron transport chain, between experiments.

In this paper, we propose a one-dimensional polarization beam splitting grating under normal incidence with excellent polarization characteristics and a high diffraction efficiency. The main structure is a double-groove slanted grating. We employed a rigorous coupled-wave analysis and simulated annealing algorithm for numerical calculations of the grating.

The physical analytical equation is always a pursuit of designing a grating for applications. We presented analytical equations to describe the diffraction process of a deep-etched fused silica grating under the second Bragg incidence by using the simplified modal method, particularly in a special case where the effective refractive index of mode 1 might be equal to mode 2. We found that the joint effect of modes 1 and 2 has no effect on the diffraction efficiency for the 0th order and the -2nd order.
