The deep reinforcement learning (DRL) has made significant progress in various simulation environments. However, applying DRL methods to real-world scenarios poses certain challenges due to limitations in visual fidelity, scene complexity, and task diversity within existing environments. View Article and Find Full Text PDFBackground: Clinical trials have shown that neoadjuvant anlotinib combined with PD-1 blockade therapy can prolong the survival of patients with driver gene negative non-small cell lung cancer (NSCLC), but some patients fail to benefit from the combination therapy.
Methods: To explore the potential drug resistance mechanism and predict the efficacy of neoadjuvant therapy in NSCLC patients, we used scRNA-seq to observe and analyze the dynamic changes of immune cells, stromal cells and cancer cells in NSCLC patients who received neoadjuvant combination therapy. We analyzed transcriptome data of ~ 47,000 single cells from 9 NSCLC patients, including 3 treatment naïve patients, 3 post-treatment patients with major pathological response (MPR), and 3 Non-MPR patients.

View Article and Find Full Text PDFThe mortality rate of oesophageal squamous cell carcinoma (ESCC) remains high, and conventional TNM systems cannot accurately predict its prognosis, thus necessitating a predictive model. In this study, a 17-gene prognosis-related gene signature (PRS) predictive model was constructed using the random survival forest algorithm as the optimal algorithm among 99 machine-learning algorithm combinations based on data from 260 patients obtained from TCGA and GEO. The PRS model consistently outperformed other clinicopathological features and previously published signatures with superior prognostic accuracy, as evidenced by the receiver operating characteristic curve, C-index and decision curve analysis in both training and validation cohorts.

Methods: Multiplex fluorescence-based immunohistochemistry was conducted to assess Siglec-15 expression and tumor-infiltrating immune cells in LUAD from Tianjin cohort, with validation cohorts Xinchao 04 and 07.
Results: This study revealed that Siglec-15 was positively correlated with CD8 T cells and tumor-associated macrophages (TAMs) infiltration, but CD8 T cells were mostly infiltrated in the stroma area, not in the tumor area.

Despite numerous studies on tissue-engineered injectable cartilage, it is still difficult to realize stable cartilage formation in preclinical large animal models because of suboptimal biocompatibility, which hinders further application in clinical settings. In this study, we proposed a novel concept of cartilage regeneration units (CRUs) based on hydrogel microcarriers for injectable cartilage regeneration in goats. To achieve this goal, hyaluronic acid (HA) was chosen as the microparticle to integrate gelatin (GT) chemical modification and a freeze-drying technology to create biocompatible and biodegradable HA-GT microcarriers with suitable mechanical strength, uniform particle size, a high swelling ratio, and cell adhesive ability.

Liquid-liquid phase separation at complex interfaces is a common phenomenon in biological systems and is also a fundamental basis to create synthetic materials in multicomponent mixtures. It remains a grand challenge to prepare anisotropic crystal superstructures with sensitive optical properties in polymer science and materials field. This study demonstrates that semicrystalline polymers develop into anisotropic hollow spherulitic crystals spontaneously at interfaces of liquid drops. In contrast to conventional spherulites with centrosymmetric optics and grain boundaries, these anisotropic spherulitic crystals have vanished boundary defects, tunable aspect ratios, and noncentrosymmetric, orientation-sensitive birefringence.
