Targeting the gut-liver axis has emerged as a promising strategy in the treatment of metabolic dysfunction-associated steatotic liver disease (MASLD), a condition that currently represents the most common cause of chronic liver disease worldwide. Within this axis, the duodenum serves not only as a site of nutrient absorption but also as a metabolic sensor capable of influencing systemic and hepatic homeostasis. We have read with great interest the recent study by Yu , investigating the effects of duodenal mucosal ablation (DMA) by irreversible electroporation in a rat model of MASLD. The authors reported remarkable reductions in hepatic lipid content, improvements in serum lipid profiles, and both structural and functional changes in the intestinal mucosa, including alterations in enteroendocrine signaling. These results corroborate the pivotal role of the gut-liver axis in the pathogenesis of MASLD and highlight the potential of minimally invasive approaches targeting the proximal intestine. In this letter, we discuss the broader implications of these findings, emphasizing the translational relevance of intestinal modulation strategies in the comprehensive treatment of MASLD.

Metaflammation, a chronic immune response triggered by metabolic dysregulation, poses significant threats to gut-liver homeostasis in aquaculture species. To understand the progression of metaflammation, it is crucial to examine the role of SOCS8 deficiency in socs8 zebrafish, as this species may serve as a disease model for metabolic disorders due to the gradual dysregulation of immunity, metabolism, and the gut microbiota observed in them. This study examines the immune-metabolic crosstalk in grass carp, subjected to soybean meal-induced enteritis, and in socs8 zebrafish under genetic and dietary stress.

Cirrhosis is the terminal stage of various chronic liver diseases, and its decompensated stage is mainly characterized by serious complications, such as hepatic encephalopathy, ascites, spontaneous bacterial peritonitis, and gastrointestinal bleeding. Gut microbial dysbiosis is prevalent in patients with cirrhosis. Considering the bidirectional regulation of the gut-liver axis, dysbiosis is closely related to the development and progression of liver cirrhosis.

The global incidence of early-onset cancer has surged by nearly 80% over the past three decades, yet the underlying causes remain poorly understood. While genetics and lifestyle are among the traditional risk factors, emerging evidence implicates the human microbiome as a potent and overlooked contributor to early tumorigenesis. Increases in the studies that are exploring the tissue-specific microbiome signatures such as the enrichment of Actinomyces and Bacteroidia in early-onset colorectal cancer, or Enterobacter and Neisseria in pancreatic tumors offer compelling evidence for age-stratified microbial contributions.

The gut-liver axis constitutes a bidirectional communication network that integrates the ecological microenvironment, metabolism, and immune signals between the gut and liver systems. Currently, there is compelling evidence indicating that the overall dysfunction of the gut-liver axis is a pivotal driver in the pathogenesis of advanced liver diseases. This review focuses on the latest research progress regarding various components of the intestinal barrier and how these components contribute to the onset and progression of cirrhosis and its complications.
