The biological activities of natural flavonoids are structure-dependent. To investigate the structure-dependent activities of neohesperidin dihydrochalcone (NHDC) and neohesperidin (NH), which have the same basic skeletal structure, we systematically compared their roles in alleviating ovalbumin-induced food allergies in mice. Our results indicate that NHDC is superior to NH in ameliorating allergic symptoms, especially in restoring T helper 1 and T helper 2 cells (Th1 and Th2, respectively)balance and inhibiting splenic NOTCH/nuclear factor kappa-B (NF-κB) activation by enhancing binding to Hes1, which was validated by molecular docking. Both compounds increased the abundance of beneficial gut microbiota (e.g., Lactobacillus). Still, the dihydrochalcone portion of NHDC possessed superior intestinal barrier repair and immunomodulatory effects, synergizing anti-inflammatory and microbiota modulatory effects. These results provide new insights into the structure-dependent activity of natural flavonoids in treating food allergies.

This research explored the effects of ginseng residue oligosaccharides (GRO-N) and ginseng polysaccharides (GP-N) on alleviating allergic rhinitis (AR). In a rat model induced by ovalbumin (OVA), both high doses of GRO-N (GRO-N-H) and GP-N (GP-N-H) significantly decreased the frequency of sneezing and rubbing behaviors in AR-affected rats. Histopathological evaluations and cytokine analyses revealed that GRO-N-H and GP-N-H notably lowered the count of goblet cells and reduced inflammatory cytokine levels in these rats.

Sucralose, a common zero-calorie sweetener, may increase food allergy (FAs) risk via immune modulation, although its mechanisms remain poorly understood. This study investigates how sucralose exacerbates allergic responses in mice that are OVA-sensitized through specific immunologic mechanisms. Sucralose exposure markedly aggravated allergic symptoms, including diarrhea, elevated serum IgE, MCP-1, and mast cell mediators, and preferentially enhanced Th2 responses without affecting Th1 or Treg cytokine pathways.

Egg proteins, particularly ovalbumin (OVA), are a major cause of food allergies, especially in children. This study investigated the impact of ultrasound treatment on the molecular structure, digestibility, and allergenicity of OVA. Enzyme-linked immunosorbent assay (ELISA) analysis demonstrated that ultrasound treatment enhanced the enzymatic hydrolysis of OVA during in vitro gastrointestinal digestion, significantly reducing its allergenicity.
