Article Abstract

Uncertainty is a central feature of the novel coronavirus era and a primary cause of stress and anxiety. Understanding the uncertainties that people are experiencing is important to provide support and help them cope; it is also a valuable opportunity to contribute to health communication theory. Guided by the Tripartite Model of Uncertainty (TMU), this project was designed to examine different forms of uncertainty that the COVID-19 pandemic created. Interviews were conducted with 62 individuals. Findings provide some support for the TMU, in that medical and personal uncertainty were salient to people's lives during the pandemic. Findings also indicated prospects for expanding the TMU to reflect (a) systems uncertainty and (b) deeply intertwined medical-social uncertainties that may be unique to this particular type of health crisis.

Mounting evidence indicates that viruses exploit elevated reactive oxygen species (ROS) levels to promote replication and pathogenesis, yet the mechanistic underpinnings of this viral strategy remain elusive for many viral systems. This study uncovers a sophisticated viral counter-defense mechanism in the Cryphonectria hypovirus 1 (CHV1)-Fusarium graminearum system, where the viral p29 protein subverts host redox homeostasis to overcome antiviral responses. That p29 directly interacts with and inhibits the enzymatic activity of fungal NAD(P)H-dependent FMN reductase 1 (FMR1), leading to increased ROS accumulation and subsequent autophagy activation is demonstrated.

Mixed chronic scrotal pain secondary to piriformis scarring treated with PRF: case report.

In the complex pathological context of mixed pain, where nociceptive, neuropathic, and nociplastic mechanisms coexist and interact, we present an innovative diagnostic and therapeutic model for refractory chronic scrotal pain (CSP) in a 49-year-old man. The pain originated from pudendal nerve entrapment secondary to piriformis scarring. Comprehensive evaluation revealed mixed pain mechanisms: neuropathic (lancinating pain, S2-S4 dermatomal hypoesthesia, and MRI-confirmed nerve compression), nociceptive (MRI-documented proven inflammation and mechanical stress exacerbation), and nociplastic (central sensitization with prolonged pain duration and psychological comorbidities).

Preeclampsia (PE) involves complex metabolic-inflammatory interactions, yet the mechanistic links among glycolysis, protein lactylation, and pyroptosis in placental pathogenesis remain undefined. In this study, we explore their tripartite relationship with PE development by combining bioinformatics analysis of PE-associated transcriptomes with experimental validation using placental tissues from PE patients and healthy controls. To elucidate the underlying mechanism, we utilize models involving hypoxic endothelial cell cultures, pharmacological glycolysis inhibition via 2-deoxyglucose, and genetic modulation of hexokinase 2 (HK2) expressions through siRNA silencing and plasmid-based overexpression.
