Article Abstract

To investigate the transcriptional changes and cell interactions following trigeminal neuralgia (TN) in different cell types in rostral ventromedial medulla (RVM). In this study, trigeminal neuropathic pain was induced in mice by ligating the left infraorbital nerve. Ten days after nerve ligation, we performed single-nucleus RNA sequencing of the RVM cells from the Sham and TN groups. We identified neurons, astrocytes, microglial cells, oligodendrocytes, oligodendrocyte progenitor cells, Purkinje cells, neuroblasts, endothelial cells and fibroblasts in RVM tissue. After analyzing the cell-type-specific transcriptional changes in the RVM following nerve ligation, we found that the number of neurons and Purkinje cells in the RVM increased. Furthermore, the differentially expressed genes (DEGs) in neurons and astrocytes between the two groups was identified. The downregulated DEGs in neurons were significantly enriched in GABAergic synapse (such as and ). The upregulated DEGs in neurons were enriched in glutamatergic synapse and voltage-gated ion channels. The downregulated DEGs in astrocytes involved in regulation of the postsynaptic membrane and synaptic membrane were enriched. Notably, the CellChat analysis highlights the Slit2-Robo 1/2 signaling pathway as a key route of communication between neurons and astrocytes after nerve ligation. This study analyzed cell-type-specific transcriptional responses to pain, and identified the possible key genes involved in TN. We inferred cell-state-specific communication in various cell types. Our findings provide potential targets, such as Slit2-Robo 1/2, Gabrg3, Gabra2, Grm4, Grik2, Cadps2 and Camk4 on therapeutic strategies for neuropathic or orofacial pain.

Objective: Diabetic foot ulcer (DFU) is a common and serious complication of diabetes, often leading to infection, amputation and poor quality of life. Bone marrow mesenchymal stem cells (BMSCs) have shown promise in treating chronic wounds, but their therapeutic efficacy is limited due to poor survival and low regenerative activity. Low-intensity pulsed ultrasound (LIUS), a non-invasive physical modality, has been shown to enhance the biological behavior of BMSCs.

Characterization of CNS Network Changes in Two Rodent Models of Chronic Pain.

Neuroimaging in rodents holds promise for advancing our understanding of the central nervous system (CNS) mechanisms that underlie chronic pain. Employing two established, but pathophysiologically distinct rodent models of chronic pain, the aim of the present study was to characterize chronic pain-related functional changes with resting-state functional magnetic resonance imaging (fMRI). In Experiment 1, we report findings from Lewis rats 3 weeks after Complete Freund's adjuvant (CFA) injection into the knee joint (n = 16) compared with the controls (n = 14).

Background: Intracranial solitary fibrous tumors (SFTs) are rare mesenchymal tumors often presenting with dural-based lesions. These tumors can exhibit aggressive characteristics with high recurrence rates and extracranial metastasis. While SFTs occasionally invade venous sinuses, cases where the tumor arises within the venous sinus are rare.

Background: Chronic constriction injury (CCI) of the sciatic nerve induces neuropathic pain, inflammation, oxidative stress, and neurodegenerative changes, impairing sensory and emotional function. While curcumin is well recognized for its anti-inflammatory and neuroprotective properties, its therapeutic use is limited by poor bioavailability. Curcumin liposomal nanoparticles (CLNs) offer improved delivery and stability.

Objectives: To clarify the role of hippocampal glutamate system in regulating HPA axis in mediating the effect of electroacupuncture (EA) at the heart meridian for improving myocardial injury in rats with acute myocardial ischemia (AMI).

Methods: Male SD rats were randomized into sham-operated group, AMI group, EA group, and L-glutamic acid+EA group (=9). Rat models of AMI were established by left descending coronary artery ligation, and EA was applied at the "Shenmen-Tongli" segment; the rats in L-glutamic acid+EA group were subjected to microinjection of L-glutamic acid into the bilateral hippocampus prior to AMI modeling and EA treatment.
