A series of hybrids (8a-h and 11a-h) containing 2,4-diphenylpyrimidine scaffold and coumarin moiety were designed and synthesized as novel focal adhesion kinase (FAK) inhibitors for the intervention of non-small-cell lung cancer (NSCLC). Most compounds effectively suppressed the proliferative of NSCLC cells, and compound 8a was identified as the most active compound with IC value of 0.28 μM in H1299 cells, superior to TAE226 (IC = 2.28 μM). In addition, 8a was also found to inhibit the invasion and migration of NSCLC cells. Furthermore, 8a exhibited potent kinase inhibitory activity of FAK (IC = 4.968 nM) with a considerable selectivity profile against various kinase families, subsequently resulting in cell cycle arrest, apoptosis- inducing as well as the decrease of MMP-2 and MMP-9 expression in H1299 cells dose-dependently. Moreover, 8a was relatively safe to mice and inhibited the growth of implanted NSCLC tumors more potently than TAE226 in mice. Therefore, 8a may be a promising candidate for the treatment of NSCLC.

We introduce a novel method using a kilohertz (kHz) amplified 800 nm laser for the first experimental confinement of microparticles within a single beam. This study demonstrates that high-energy kHz pulses can confine 1-μm-radius polystyrene beads in water within ∼26 μm. This approach utilizes the unique properties of high-energy pulsed lasers, distinct from continuous-wave and megahertz pulsed lasers traditionally used in optical trapping.

Modulating cell endocytosis activity to reduce host susceptibility to virus represents a promising strategy for antiviral drug development. In this study, we reveal that lactate transporter SLC16A3 is a critical host factor for reducing diverse virus invasion. By performing metabolomics, proteomics, and thermal proteome profiling experiments, AP1G1, a pivotal protein involved in cellular endocytosis, was indiscriminately screened as a chaperone of SLC16A3.

Methods: To address these limitations, a lightweight rice leaf spot segmentation model (MV3L-MSDE-PGFF-CA-DeepLabv3+, MMPC-DeepLabv3+) was developed for three common rice leaf diseases: rice blast, brown spot and bacterial leaf blight.

Stage IV prostate cancer (PCa) refers to a disease that has metastasized beyond the prostate gland to distant sites, such as bones, visceral organs, or non-regional lymph nodes. While early attempts at curative therapy were occasionally made in oligometastatic cases, current guidelines uniformly recommend palliative-intent management once true metastatic spread is confirmed. Over the past decade, treatment paradigms have shifted from androgen deprivation therapy (ADT) monotherapy to earlier intensification with combination regimens including chemo-hormonal therapy and next-generation hormonal agents to improve survival and quality of life (QoL).
