Elemene is widely recognized as an effective anti-cancer compound and is routinely administered in Chinese clinical settings for the management of several solid tumors, including non-small cell lung cancer (NSCLC). However, its detailed molecular mechanism has not been adequately demonstrated. In this research, it was demonstrated that elemene effectively curtailed NSCLC growth in the patient-derived xenograft (PDX) model. Mechanistically, employing high-throughput screening techniques and subsequent biochemical validations such as microscale thermophoresis (MST), microRNA-145-5p (miR-145-5p) was pinpointed as a critical target through which elemene exerts its anti-tumor effects. Interestingly, elemene serves as a binding stabilizer for miR-145-5p, demonstrating a strong binding affinity (dissociation constant ( ) = 0.39 ± 0.17 μg/mL) and preventing its degradation both and , while not interfering with the synthesis of the primary microRNA transcripts (pri-miRNAs) and precursor miRNAs (pre-miRNAs). The stabilization of miR-145-5p by elemene resulted in an increased level of this miRNA, subsequently suppressing NSCLC progression through the miR-145-5p/mitogen-activated protein kinase kinase kinase 3 (MAP3K3)/nuclear factor kappaB (NF-κB) pathway. Our findings provide a new perspective on revealing the interaction patterns between clinical anti-tumor drugs and miRNAs.

Parvalbumin (PV), a thermostable and digestion-resistant fish allergen, has been shown to retain its allergenic potential following traditional treatments, thus posing a persistent allergic risk. The study investigated the digestive kinetics and IgE immunoreactivity of Trachinotus ovatus PV, a major fish allergen, under different treatments (untreated; DPCD treatment-15 MPa, 30 min, 50 °C; heat treatment), to evaluate its allergenic potential alterations. The development of antiviral nanotherapeutics remains a formidable challenge due to the structural diversity and rapid evolution of viral surface glycoconjugates. Here, we report a rationally engineered multivalent Galectin-1 (Gal-1) nanoplatform based on 13-nm gold nanoparticles (AuNPs) for high-affinity glycan targeting and therapeutic inhibition of influenza virus. By leveraging site-specific conjugation and molecular orientation control, the AuNP/Gal-1 nanocomplex maximizes the exposure of carbohydrate recognition domains (CRDs) while preserving Gal-1's tertiary structure, as confirmed by molecular dynamics simulations and spectroscopic analyses.

The voltage-dependent anion channel (VDAC) family proteins can be subdivided into three isoforms: VDAC1, VDAC2, and VDAC3. As core channels of the mitochondrial outer membrane, these proteins exhibit paradoxical regulatory roles in cancer development.
