Prognostic biomarkers have been widely studied in COVID-19, but their levels may be influenced by treatment strategies. This study examined plasma biomarkers and proteomic survival prediction in two unvaccinated hospitalized COVID-19 cohorts receiving different treatments. In a derivation cohort ( = 126) from early 2020, we performed plasma proteomic profiling and evaluated innate and complement system immune markers. A proteomic model based on differentially expressed proteins predicted 30-day mortality with an area under the curve (AUC) of 0.81. The model was tested in a validation cohort ( = 80) from late 2020, where patients received remdesivir and dexamethasone, and performed with an AUC of 0.75. Biomarker levels varied considerably between cohorts, sometimes in opposite directions, highlighting the impact of treatment regimens on biomarker expression. These findings underscore the need to account for treatment effects when developing prognostic models, as treatment differences may limit their generalizability across populations.

Objectives: This study aims to assess periodontal and biochemical parameters and evaluate the salivary Protectin D1 levels in periodontitis patients with and without metabolic syndrome after non-surgical periodontal therapy.
Materials And Methods: Forty patients were categorized into two groups: 20 patients in Group P (systemically healthy patients with stage II/III grade B periodontitis) and 20 patients in Group P+MS (patients with stage II/III grade B periodontitis and metabolic syndrome). Parameters including age, gender, height, weight, body mass index, waist circumference, socio-economic status, oral hygiene index (OHI), modified gingival index (MGI), probing pocket depth, clinical attachment levels, fasting blood glucose, HDL-c, total triglycerides, and blood pressure were recorded.

Infection with SARS-CoV-2 elevates the expression of cytokines, resulting in a cytokine storm that serves as the primary factor for severe illness and mortality; however, effective markers for predicting disease severity and preventing are lacking. Thus, we investigated the association between serum levels of nerve injury-induced protein 1 (Ninj1), a mediator of plasma membrane rupture, and the extent of lung damage in COVID-19 patients was examined to anticipate the severity of SARS-CoV-2 infection. Wendan Decoction (WDD), a classic formula for the expectoration of phlegm, has been acknowledged for the management of chronic obstructive pulmonary disease (COPD). Nevertheless, the therapeutic components and mechanism of WDD in COPD remain elusive. Hence, an analytical strategy for extensive investigation of the constituents in WDD combining UFLC-ESI-Q/TOF-MS and subsequent network pharmacology was conducted, and the appropriate preparation of bio-samples was involved with mechanistic bioanalysis including multi-technique metabolomics and molecular biological means, that pivotal factors such as PTGS2, arachidonic acid (AA) metabolism, and PI3K-AKT signaling pathway were recommended as potential central elements for the anti-COPD effects.
