Article Abstract

HSA mice are the most broadly used animal model for studying myotonic dystrophy type I (DM1). However, so far, HSA preclinical studies have often excluded female mice or failed to document the biological sex of the animals. This leaves an unwanted knowledge gap concerning the differential development of DM1 in males and females, particularly considering that the disease has a different clinical presentation in men and women. Here we compared typical functional measurements, histological features, molecular phenotypes and biochemical plasma profiles in the muscles of male and female HSA mice in search of any significant between-sex differences that could justify this exclusion of female mice in HSA studies and, critically, in candidate therapy assays performed with this model. We found no fundamental differences between HSA males and females during disease development. Both sexes presented comparable functional and tissue phenotypes, with similar molecular muscle profiles. The only sex differences and significant interactions observed were in plasma biochemical parameters, which are also intrinsically variable in patients with DM1. In addition, we tested the influence of age on these measurements. We therefore suggest including female HSA mice in regular DM1 studies, and recommend documenting the sex of animals, especially in studies focusing on metabolic alterations. This will allow researchers to detect and report any potential differences between male and female HSA mice, especially regarding the efficacy of experimental treatments that could be relevant to patients with DM1.

Objective: CircRNAs are involved in cancer progression. However, their role in immune escape in non-small cell lung cancer (NSCLC) remains poorly understood.

Methods: This study employed RIP-seq for the targeted enrichment of circRNAs, followed by Western blotting and RT-qPCR to confirm their expression.

Various cancer therapeutic strategies have been designed for targeting tumor-associated macrophages (TAMs), but TAM reprogramming-based monotherapy is often clinically hindered, likely due to the lack of a coordinated platform to initiate T cell-mediated immunity. Herein, we fabricated reactive oxygen species (ROS)-responsive human serum albumin (HSA)-based nanoparticles (PEG/IL12-IA NPs) consisting of indocyanine green (ICG), arginine (Arg), and interleukin 12 (IL12). Upon laser irradiation, the nanoparticles were found to be able to dissociate, thus facilitating the release of IL12.

Objectives: Circular RNAs (circRNAs) are a class of non-coding RNAs with diverse pathophysiological functions. However, the functional roles and molecular mechanisms of circRNAs in lung adenocarcinoma (LUAD) remain to be further elucidated.

Methods: The expression levels of Circ_0007552 (Circ_RILPL1), miR-7974, and BRCA1-associated protein 1 (BAP1) mRNA in LUAD tissues and cells were detected by quantitative real-time PCR (qRT-PCR).

Fatigue is a common but poorly understood issue in type 2 diabetes (T2DM) that affects quality of life. Although ceRNA networks regulate disease progression, their role in T2DM-related fatigue (F-T2DM) is unclear. This study developed a circRNA-mediated ceRNA network to uncover the molecular interactions causing fatigue in F-T2DM.

circHMGCS1 Promotes the Progression of NSCLC by Regulating the HuR/SFPQ/TNF Pathway.

Among lung cancers, non-small-cell lung cancer (NSCLC) is the most common type. Evidence demonstrates that abnormal expression of circular RNAs (circRNAs) is associated with cancer progression through interactions with RNA-binding proteins (RBPs). Therefore, further elucidation of the role of circular RNAs may provide new therapeutic targets for NSCLC patients.
