Article Abstract

Tissue-resident memory T cells (T) can be induced by infection and vaccination, and play a key role in maintaining long-term protective immunity against mucosal pathogens. Our studies explored the key factors and mechanisms affecting the differentiation, maturation, and stable residence of gastric epithelial CD4 T induced by Helicobacter pylori (Hp) vaccine and optimized Hp vaccination to promote the generation and residence of T. Cluster of differentiation (CD)38 regulated mitochondrial activity and enhanced transforming growth factor-β signal transduction to promote the differentiation and residence of gastric epithelial CD4 T by mediating the expression of CD105. Extracellular nucleotides influenced the long-term maintenance of T in gastric epithelium by the P2X7 receptor (P2RX7). Vitamin D3 and Gram-positive enhancer matrix (GEM) particles as immune adjuvants combined with Hp vaccination promoted the production of CD69CD103CD4 T.

Mucin 6 (MUC6), primarily expressed in the gastrointestinal (GI) epithelium, is a member of the mucin family characterized by a protein backbone with extensive glycosylation, playing a crucial role in preserving epithelial barrier integrity. Accumulating evidence indicates that MUC6 glycosylation contributes significantly to cancer development, diagnosis, therapy and prognosis‑particularly in GI malignancies such as gastric, pancreatic and colorectal cancers. A 68-year-old man came to evaluate a gastric polyp discovered during a routine gastroscopy. After endoscopic mucosal resection, pathological findings confirmed that it's a hyperplastic polyp. Notably, squamous metaplasia was observed within the hyperplastic polyp, and both squamous and glandular epithelium exhibited high-grade intraepithelial neoplasia.
