Article Abstract

is a causal agent of sugarcane Pokkah boeng, an important fungal disease that causes a considerable reduction in yield and sugar content in susceptible varieties of sugarcane worldwide. Despite its importance, the fungal factors that regulate the virulence of this pathogen remain largely unknown. In our previous study, mapping of an insertional mutant defect in virulence resulted in the identification of a cutinase G-box binding protein gene, designated that encodes a C2H2-type transcription factor (TF). FsCGBP was shown to localize in the nuclei, and the transcript level of was significantly upregulated during the infection process or in response to abiotic stresses. Deletion or silencing of FsCGBP resulted in a reduction in mycelial growth, conidial production, and virulence and a delay in conidial germination in the . . Cutinase genes , , and and the mitogen-activated protein kinase (MAPK) genes , and which were significantly downregulated in Δ. Except for , all of these genes were found to be transcriptionally activated by FsCGBP using the yeast one-hybrid system in vitro. The deletion of individual cutinase genes did not result in any of the phenotypes exhibited in the Δ mutant, except for cutinase activity. However, disruption of the MAPK pathway upon deletion of or resulted in phenotypes similar to those of the Δ mutant. The above results suggest that FsCGBP functions by regulating the MAPK pathway and cutinase genes, providing new insights into the mechanism of virulence regulation in .

Poly (butylene adipate-co-terephthalate) (PBAT) is a biodegradable polyester widely used in agriculture and packaging. However, its slow decomposition under natural conditions raises environmental concerns. In this study, we explored strategies to enhance PBAT degradation by Purpureocillium lilacinum strain BA1S.

T7 RNA polymerase (T7RNAP), orthogonal to the T7 promoter, is a powerful tool in engineered that enables the production of many different harsh enzymes. Still, it requires precise control, particularly when expressing toxic proteins. The optimized strategy for the interconnected processes of transcription (TX), translation (TL), and protein folding (FD) in the T7 system is still not well understood.

Fishscale bamboo rhombic-spot, caused by , poses a significant threat to in Sichuan province. Based on genomic analysis, four cutinase genes, NsCut1-NsCut4, were identified, cloned, and functionally validated. Bioinformatics analyses revealed that the proteins encoded by these genes possess secretory functions, lack transmembrane domains, and contain conserved cutinase domains highly homologous to those in other fungi.

Advanced Materials Program, Department of Biological Engineering, College of Engineering, Konkuk University, Seoul, Republic of Korea; Institute for Ubiquitous Information Technology and Applications, Konkuk University, Seoul, Republic of Korea. Electronic address:

With the increase in plastic production, efficient and timely plastic degradation are urgently needed. In that point, biodegradable plastics have attracted attention as potential solutions for environmental pollution of plastics. However, finding of superior degrading strains and enzymes such as esterase, cutinase, and triacylglycerol lipase (TGL) of bioplastic are still needed together with the efficient secretion systems of degrading enzymes.
