Article Abstract

Cucumber (Cucumis sativus, Cs) tendrils are slender vegetative organs that typically require manual removal to ensure orderly growth during greenhouse cultivation. Here, we identified cucumber tendril-less (tl), a Tnt1 retrotransposon-induced insertion mutant lacking tendrils. Map-based cloning identified the mutated gene, CsaV3_3G003590, which we designated as CsTL, which is homologous to Arabidopsis thaliana LATERAL SUPPRESSOR (AtLAS). Knocking out CsTL repressed tendril formation but did not affect branch initiation, whereas overexpression (OE) of CsTL resulted in the formation of two or more tendrils in one leaf axil. Although expression of two cucumber genes regulating tendril formation, Tendril (CsTEN) and Unusual Floral Organs (CsUFO), was significantly decreased in CsTL knockout lines, these two genes were not direct downstream targets of CsTL. Instead, CsTL physically interacted with CsTEN, an interaction that further enhanced CsTEN-mediated expression of CsUFO. In Arabidopsis, the CsTL homolog AtLAS acts upstream of REVOLUTA (REV) to regulate branch initiation. Knocking out cucumber CsREV inhibited branch formation without affecting tendril initiation. Furthermore, genomic regions containing CsTL and AtLAS were not syntenic between the cucumber and Arabidopsis genomes, whereas REV orthologs were found on a shared syntenic block. Our results revealed not only that cucumber CsTL possesses a divergent function in promoting tendril formation but also that CsREV retains its conserved function in shoot branching.

MADS-box transcription factors play a crucial role in plant growth and development. Although previous genome-wide analyses have investigated the MADS-box family in cucumber, this study provides the first comprehensive reannotation of the gene family in using updated Cucurbitaceae genome data, offering novel insights into the gene family's evolution and functional diversity. The results show that a total of 48 genes were identified in the V3 version of cucumber, while 3 of the 43 genes identified in the V1 version were duplicated.

One of Earth's largest carbon fluxes is driven by particles made from photosynthetically fixed matter, which aggregate and sink into the deep ocean. While biodegradation is known to reduce this vertical flux, the biophysical processes that control particle sinking speed are not well understood. Here, we use a vertical millifluidic column to video-track single particles and find that biogels scavenged by particles during sinking significantly reduce the particles' sinking speed, slowing them by up to 45% within one day.

Nano-tendril bundles (NTBs) are clusters of fuzzy nanostructures that form on tungsten surfaces when they are exposed to helium plasmas under specific conditions, reaching lengths of over 100 µm. Tungsten samples with NTBs and fuzz were exposed to high-density hydrogen plasmas in the Upgraded Pilot-PSI linear plasma device to investigate their response, with a particular focus on unipolar arc ignition and hot spot formation. Although NTBs are prone to arcing due to their high field electron emission characteristics, unipolar arcs have never occurred on NTBs.
