Article Abstract

Targeting Protein for Xenopus Kinesin Like Protein 2 (TPX2) serves as a microtubule associated protein for the regulation of spindle assembly and tumorigenesis. We aim to investigate the prognostic and immunological role of TPX2 in pan-cancer. TCGA database, Tumor Immune Single-cell Hub (TISCH), and Human Protein Atlas (HPA) were retrieved to evaluate the expression pattern of TPX2 as well as its diagnostic and prognostic value in solid tumors. Genomic alterations of TPX2 were assessed with cBioPortal database. experiments in lung adenocarcinoma (LUAD) were performed to confirm the potential role of TPX2. Overexpression of TPX2 was found in 22 types of cancers, and was positively related with copy number variations (CNV) and negative with methylation. Up-regulated TPX2 could predict worse outcomes in the majority of cancers. Single-cell analysis revealed that TPX2 was mainly distributed in malignant cells (especially in glioma) and proliferating T cells. Genomic alteration of TPX2 was common in different types of tumors, while with prognostic value in two types of cancers. Additionally, significant correlations were found between TPX2 expression and tumor microenvironment (including stromal cells and immune cells) as well as immune related genes across cancer types. Drug sensitivity analysis revealed that TPX2 could predict response to chemotherapy and immunotherapy. Functional analyses demonstrated close relationship of TPX2 with immune function and malignant phenotypes. Finally, it was confirmed that knockdown of TPX2 could reduce proliferation and migration ability of LUAD cells. In summary, TPX2 could serve as a diagnostic and prognostic biomarker and a potential immunotherapy marker.

The RNA-binding protein OsDMCK1 is required for meiotic cytokinesis by regulating cell plate formation in rice.

Plant cytokinesis is distinguished from animal cytokinesis by the formation of a cell plate between dividing cells. While meiotic cytokinesis involves two successive nuclear divisions with distinct regulatory mechanisms from mitosis, the underlying mechanisms remain poorly understood. In this study, we identified OsDMCK1, a novel rice RNA-binding protein essential for male fertility.

As natural chemicals may have fewer side effects than conventional medicines, their usage in cancer treatment is becoming more popular. Nanoformulations have been the focus of recent developments to increase the anticancer potential of phytocompounds or pure isolated components. To evaluate the anticancer potential of silver nitrate nanoparticles (AgNPs), essential oil (EO) from was extracted, a terpene compound was isolated and incensole acetate (IA)-mediated silver nanoparticles (IA-AgNPs) were synthesized.

Background: Diminished ovarian reserve (DOR) becomes more common with age, and hypoxia is a key cause of apoptosis in ovarian granulosa cells. This study investigated the genetic links between hypoxia and DOR.

Methods: The GSE87201 dataset for DOR was sourced from Gene Expression Omnibus database, normalized for common differentially expressed genes (Co-DEGs), and identified Hypoxia-related differentially expressed genes (HRDEGs) GeneCards; Receiver Operating Characteristic (ROC) curves evaluated HRDEGs' diagnostic value, and protein-protein interaction networks were visualized with STRING and Cytoscape.

Previous studies revealed the presence of several tight junction (TJ) proteins in the centrosome and their interaction with various centriolar proteins, prompting us to analyze whether this also applies to the TJ protein ZO-2. Here, we found that ZO-2 colocalizes with CEP164 in the distal appendage of the mother centriole and is also present in the pericentriolar region, mitotic spindle poles, the basal body of primary cilia, and the tail of spermatozoa. The absence of ZO-2 altered the cellular content of centriolar proteins CEP164, centriolin, and CEP135, but did not change the morphology of centrioles.

KIF16B is a member of the kinesin-3 family of motor proteins, which facilitates processes such as vesicle transport, microtubule dynamics, and organelle function during mitosis. In this study, we explored the role of KIF16B in meiosis. Our findings indicate that KIF16B is involved in the meiotic G2-M transition and spindle assembly in oocytes.
